ABSTRACT A quarter of a century ago, Liise-anne Pirofski and Arturo Casadevall shared their concepts microbial pathogenesis through the lens damage-response framework (DRF), which characterizes disease by assessing dynamic interactions between host pathogen as reflected damage readout. This has evolved to be powerful tool for understanding biology complex infectious diseases, analyzing emerging reemerging microbes, developing therapeutic approaches combat infections. The DRF is also frequently used explain research at scientific meetings teach diverse learners. mGem reviews how came provides an overview it used. Without doubt, community will continue leverage advance innovate approaches, especially important new diseases threaten global health.

Thermal adaptability is a crucial characteristic for mammalian pathogenic fungi that originally inhabit natural ecosystems. Thermally dimorphic have evolved unique ability to respond host body temperature by shifting from mycelia yeast. The high similarity of protein-coding genes between these and their relatives suggests the indispensable but often overlooked roles non-coding elements in fungal thermal adaptation. Here, we systematically delineated landscape full-length circRNAs both mycelial yeast conditions Talaromyces marneffei, typical thermally fungus causing fatal Talaromycosis, optimizing an integrative pipeline circRNA detection utilizing next- third-generation sequencing. We found T. marneffei demonstrated features such as shorter length, lower abundance, circularization-biased splicing. then identified validated circDS-1, independent its parental gene, promotes hyphae-to-yeast transition, maintains morphology, involved virulence regulation. Further analysis experiments among confirmed generation circDS-1 driven marneffei-specific region flanking intron circDS-1. Together, our findings not only provide fresh insights into role adaptation also reveal novel molecular mechanism adaptive evolution functional derived intronic mutations.

Talaromyces marneffei is an emerging pathogen, and the number of infections in HIV-negative individuals increasing. In individuals, talaromycosis usually misdiagnosed as another disease, especially tuberculosis (TB). We retrospectively extracted clinical data patients with infection from 2018 to 2023, analyzed differences between TB attempted establish differential diagnosis models utilizing prediction for these two diseases. Overall, 718 patients, including 137 581 pulmonary (PTB), were enrolled this study. According multivariate analysis, age > 65 years, expectoration, PLT count independent predictors TB. Fever, chest pain, gasping, rash, lymphadenectasis, osteolysis, Neu count, EOS ALB talaromycosis. Receiver operating characteristic (ROC) curve analysis training set showed that area under (AUC) (95 % CI) model based on logistic regression was 0.918 (0.884-0.953). The verified validation set. ROC AUC 0.900 (0.841-0.959). These new can calculate probability either or tuberculosis.

To explore the clinical and chest CT features of Talaromycosis marneffei (TSM), to compare differences between anti‑IFN‑γ autoantibodies‑positive HIV-positive cases. Clinical data images 54 HIV-negative patients with Talaromyces (TM) infection positive anti-interferon-γ (anti-IFN-γ) autoantibody were retrospectively analyzed. Ninety-three TM during same period included as controls. Differences groups compared by two-sample t-test Mann-Whitney U test, respectively. The time from symptom onset diagnosis TSM anti-IFN-γ autoantibodies was significantly longer than that controls (P < 0.001). BALF/sputum lymph nodes better specimens for detecting in autoantibody-positive group, while more often isolated or detected blood samples other group. Patients tended present cough, expectoration, fever. Compared controls, antibody-positive had a higher incidence shortness breath, skin lesions, joint pain, high CD4/CD8 ratio 0.05). There only 1 6 cases negative lung involvements scans infection, In remaining patients, manifestations diverse, mainly presenting fibrous cord‑like node enlargement, sporadic nodules. incidences patchy consolidation, air bronchogram, pleural effusion thickening those Anti-IFN-γ antibody showed diagnostic interval. which different two groups. imaging are characteristic.
