Thermal adaptability is a crucial characteristic for mammalian pathogenic fungi that originally inhabit natural ecosystems. Thermally dimorphic have evolved unique ability to respond host body temperature by shifting from mycelia yeast. The high similarity of protein-coding genes between these and their relatives suggests the indispensable but often overlooked roles non-coding elements in fungal thermal adaptation. Here, we systematically delineated landscape full-length circRNAs both mycelial yeast conditions Talaromyces marneffei, typical thermally fungus causing fatal Talaromycosis, optimizing an integrative pipeline circRNA detection utilizing next- third-generation sequencing. We found T. marneffei demonstrated features such as shorter length, lower abundance, circularization-biased splicing. then identified validated circDS-1, independent its parental gene, promotes hyphae-to-yeast transition, maintains morphology, involved virulence regulation. Talaromyces marneffei is an emerging pathogen, and the number of infections in HIV-negative individuals increasing. In individuals, talaromycosis usually misdiagnosed as another disease, especially tuberculosis (TB). We retrospectively extracted clinical data patients with infection from 2018 to 2023, analyzed differences between TB attempted establish differential diagnosis models utilizing prediction for these two diseases. Overall, 718 patients, including 137 581 pulmonary (PTB), were enrolled this study. According multivariate analysis, age > 65 years, expectoration, PLT count independent predictors TB. To explore the clinical and chest CT features of Talaromycosis marneffei (TSM), to compare differences between anti‑IFN‑γ autoantibodies‑positive HIV-positive cases. Clinical data images 54 HIV-negative patients with Talaromyces (TM) infection positive anti-interferon-γ (anti-IFN-γ) autoantibody were retrospectively analyzed. Ninety-three TM during same period included as controls. Differences groups compared by two-sample t-test Mann-Whitney U test, respectively. The time from symptom onset diagnosis TSM anti-IFN-γ autoantibodies was significantly longer than that controls (P < 0.001).
