The DiEt Rheumatoid Arthritis (MADEIRA) was a single-blind (statistician), two-arm randomized clinical trial, investigating effects of 12-week lifestyle intervention, including personalized isocaloric MD plan with promotion physical (PA), supported through decision support systems (CDSS) platform, versus usual care in women RA. Forty adult RA on remission were randomly allocated (1:1 ratio) either intervention or control arm. group received plans consultation improving PA levels, whereas controls given generic dietary advice, based National Dietary Guidelines. primary outcome that difference secondary outcomes included change (DAS28), anthropometric indices (BodPod), intake, PA, vitamin D concentrations, blood lipid profiles after 12 weeks from initiation trial. Rheumatoid arthritis (RA) is a systemic autoimmune disease characterized by synovial inflammation, cartilage destruction, pannus formation and bone erosion. Various immune cells, including macrophages, are involved in RA pathogenesis. The heterogeneity plasticity of macrophages render them pivotal regulators both the induction resolution inflammatory response. Predominantly, two different phenotypes have been identified: classically activated M1 exacerbate inflammation via production cytokines, chemokines other mediators, while alternatively M2 inhibit facilitate tissue repair. An imbalance M1/M2 macrophage ratio critical during initiation progression RA. Macrophage polarization modulated various transcription factors, epigenetic elements metabolic reprogramming. Curcumin, an active component turmeric, exhibits potent immunomodulatory effects administered treatment multiple diseases, regulation subsequent cytokine as well migration mechanisms underlying therapeutic effect curcumin on In this review, we summarize which modulates function context to provide evidence for clinical application treatment.

Studies have noted the connection between Mycobacterium avium subspecies paratuberculosis (MAP) and autoimmunity. MAP is an intracellular pathogen that infects multiplies in macrophages. We compared RA patients (n = 100) control subjects (CS) assessing specific immune responses (the antigen) indirect ELISA method. Results showed significant difference levels CS, having median OD 0.4645 0.1372 CS. Antibodies against were present 95% 16% CS (AUC 0.9163, p 0.0001). A BSTRACT Patients diagnosed with rheumatoid arthritis have a heightened susceptibility to infections, which may lead higher rates of illness and death. The arise from the itself, causes changes in body’s innate cellular defense mechanisms, or medications used manage condition. precise level risk for infections associated traditional disease-modifying anti-rheumatic drugs has not been fully elucidated. This review aimed To investigate type use antibiotics among patients arthritis. An electronic literature search was conducted using MEDLINE database, indicated keywords: antibiotics, use, patients, rheumatoid, identify relevant information, limited articles published between 2017 2024. researchers suitable terms on Google Scholar discover examine scholarly articles. selection determined by several inclusion criteria. research included publications that were study organized into many sections, each including particular categories within analysis section.we reportrd : Within developing age focused synthetic treatments RA, severe persist as primary consequence long-term treatment. Abstract Introduction/objective Janus kinase (JAK) inhibitors have expanded treatment options for rheumatoid arthritis (RA), particularly patients unresponsive to traditional disease-modifying antirheumatic drugs (DMARDs). However, safety concerns necessitate a thorough post-market evaluation. This study is aimed at comparing the profiles of tofacitinib, baricitinib, and upadacitinib using adverse event (AE) reports from FDA Adverse Event Reporting System (FAERS). Methods A retrospective disproportionality analysis was performed FAERS data 2012 2022. The AE were categorized into cardiovascular, cancer, respiratory, gastrointestinal, musculoskeletal, arthralgia-related events. Proportional reporting ratio (PRR) odds ratios (RORs) with 95% confidence intervals (CIs) calculated identify significant signals. Results Of 273,657 reports, tofacitinib had most (227,144), increased musculoskeletal-related events (ROR = 1.53, CI 1.49–1.57) reduced cancer risk 0.44, 0.41–0.47). Baricitinib (9305 reports) showed highest cardiovascular 1.63, 1.50–1.78) 2.17, 1.83–2.58). Upadacitinib (37,208 elevated risks respiratory 2.04, 1.88–2.21) 2.24, 2.05–2.43). Conclusion distinct these JAK suggest that baricitinib poses higher risks, whereas increases gastrointestinal Tofacitinib may be safer history but requires monitoring musculoskeletal AEs. Personalized assessments are critical safe use inhibitors. Key Points • provides comprehensive assessment three inhibitors—tofacitinib, upadacitinib—using Distinct identified, showing while posed an demonstrated lower than other associated more These findings emphasize importance personalized vigilant when prescribing arthritis.

In recent years, a possible association of lifestyle factors with rheumatoid arthritis (RA) has attracted increasing public interest. The aim this review is to provide an overview the extent and limitations current evidence regarding RA. PubMed medical database was screened for epidemiological prospective studies investigating contribution development course disease. Large have identified smoking, unhealthy diet adiposity, as well low educational level socioeconomic status, that increase incidence addition, several habits influence response RA antirheumatic drugs. Among others, obesity poor physical activity are associated worse treatment outcome. Methodological problems often impair firm conclusions respect causal role these in risk However, sufficient recommend healthy diet, prevention obesity, cessation smoking maintenance high support effectivity modern medication.

Lifestyle modifications in preclinical Rheumatoid Arthritis (RA) could delay the ongoing pathogenic immune processes and potentially prevent its onset. Physical exercise (PE) benefits RA patients; however, impact reducing risk of developing has scarcely been studied. The objective was to describe effects low-intensity PE applied at disease’s phase on joints DBA/1 mice with collagen-induced arthritis (CIA). Twelve CIA were randomly distributed into two groups: CIA-Ex group, which undertook treadmill PE, CIA-NoEx, not exercised. evaluated through clinical, histological, transcriptomics, immunodetection analyses mice’s hind paws. group showed lower joint inflammation damage a decreased expression RA-related genes (Tnf Il2, Il10, Il12a, IL23a, Tgfb1) signaling pathways (Cytokines, Chemokines, JAK-STAT, MAPK, NF-kappa B, TNF, TGF-beta). TNF-α by inflamed joints. Low-intensity pre-arthritic reduced severity down-expression proinflammatory proteins. Knowledge underlying mechanisms is still needed.

Rheumatoid arthritis (RA) is a chronic inflammatory disease that affects the joints. The multifactorial etiopathogenesis of RA has been heavily investigated, but still only partially understood. Diet can represent both risk factor and protective factor, based on some evidence suggests specific properties certain foods their ability to increase/reduce inflammation. To date, studies done this topic provide discordant results are heterogeneous in terms design cohort size. In work, we investigated for first time relationship between nutrition onset using sample size about half million subjects from one largest publicly available biobanks UK biobank. Results showed oily fish, alcohol, coffee breakfast cereals have roles RA; whereas, tea increase RA. conclusion, obtained confirm diet plays key RA, either by promoting or preventing development. Future research should focus unravelling effects dietary habits immune-mediated diseases establish better preventive strategies.
