Diabetic foot ulcers (DFU) are a major complication of diabetes mellitus and public health concern worldwide. The ability P. aeruginosa to form biofilms is key factor responsible for the chronicity diabetic infections (DFIs) frequently associated with presence persister cells. These subpopulation phenotypic variants highly tolerant antibiotics which new therapeutic alternatives urgently needed, such as those based on antimicrobial peptides. This study aimed evaluate inhibitory effect nisin Z DFI persisters. To induce development state in both planktonic suspensions biofilms, isolates were exposed carbonyl cyanide m-chlorophenylhydrazone (CCCP) ciprofloxacin, respectively. After RNA extraction from CCCP-induced persisters, transcriptome analysis was performed differential gene expression between control, cells Z. Nisin presented high against but unable eradicate them when present established biofilms. Transcriptome revealed that persistence downregulation genes related metabolic processes, cell wall synthesis, dysregulation stress response biofilm formation. treatment, some transcriptomic changes induced by reversed. In conclusion, could be considered potential complementary therapy treating DFI, it should applied an early treatment or after wound debridement.

Abstract Background The increase in the resistance of bacterial strains to antibiotics has led research into bactericidal potential non-antibiotic compounds. This study aimed evaluate vitro antibacterial/ antibiofilm properties nisin and selenium encapsulated thiolated chitosan nanoparticles (N/Se@TCsNPs) against prevalent enteric pathogens including standard isolates Vibrio (V.) cholerae O1 El Tor ATCC 14,035, Campylobacter (C.) jejuni 29,428, Salmonella (S.) enterica subsp. 19,430, Shigella dysenteriae PTCC 1188, Escherichia (E.) coli O157:H7 25,922, Listeria (L.) monocytogenes 19,115, Staphylococcus aureus 29,733. Methods synthesis comprehensive analysis N/Se@TCsNPs have been completed. Antibacterial capabilities were evaluated through broth microdilution crystal violet assays. Furthermore, included examining cytotoxic effects on Caco-2 cells exploring immunomodulatory N/Se@TCsNPs. assessing levels both pro-inflammatory (IL-6 TNFα) anti-inflammatory (IL-10 TGFβ) cytokines determining gene expression TLR2 TLR4 . Results showed an average diameter 136.26 ± 43.17 nm a zeta 0.27 0.07 mV. FTIR spectroscopy validated structural features Scanning electron microscopy (SEM) images confirmed their spherical shape uniform distribution. Bacterial keratitis (BK) causes visual morbidity/blindness if not treated effectively. Here, ciprofloxacin (CIP)-loaded nanoparticles (NPs) using glycol chitosan (GC) and poly(lactic acid) (PLA) conjugate at three different ratios (CIP@GC(PLA) NPs (1:1,5,15)) were fabricated. CIP@GC(PLA) (1:1) more effective than other tested ratios, indicating the importance of optimal hydrophobic/hydrophilic balance for corneal penetration preventing bacterial invasion. The realized highest association with human epithelial cells, which nonirritant to hen's egg-chorioallantoic membrane test (HET-CAM test) demonstrated significant antibacterial response in vitro minimum inhibitory, bactericidal, live-dead zone inhibition, biofilm inhibition assays against keratitis-inducing pathogen Pseudomonas aeruginosa. antiquorum sensing activity GC has been explored first time. disrupted quorum by inhibiting production virulence factors, including acyl homoserine lactones, pyocyanin, motility, caused downregulation associated genes. Pseudomonas aeruginosa is recognized globally as an opportunistic pathogen of considerable concern due to its high virulence and pathogenicity, especially in immunocompromised individuals. While research has identified several endogenous quorum sensing (QS) signaling molecules that enhance the pathogenicity P. aeruginosa, investigations on exogenous QS or modulating factors remain limited. This study found dopamine serves molecule factor PAO1, enhancing production biofilms. Compared control group, treatment with 40 μM resulted a 33.1% increase biofilm formation, 68.1% swimming mobility, 63.1% swarming 147.2% 3-oxo-C12-HSL, 50.5%, 28.5%, 27.0%, 33.2% increases alginate, rhamnolipids, protease, pyocyanin, respectively. further explored mechanism regulating formation PAO1 through transcriptome metabolome. Transcriptomic analysis showed promoted expression genes psl, alg, lasA, rhlABC, rml, phz PAO1. Metabolomic revealed changes concentrations tryptophan, pyruvate, ethanolamine, glycine, 3-hydroxybutyric acid, alizarin. Furthermore, KEGG enrichment altered metabolites indicated enhanced phenylalanine, tyrosine, tryptophan The results this will contribute development novel advance our understanding interactions between host environment.

Bacterial biofilm is an emerging form of life that involves cell populations living embedded in a self-produced matrix extracellular polymeric substances (EPS). Currently, little known about the molecular mechanisms Bifidobacterium formation. We used fermentation system to preparation biofilms on wheat fibers, and multi-omics analysis both B. pseudocatenulatum planktonic cells were performed identify genes metabolites involved The average diameter fibers was around 50 μm, while particle culture over 260 μm at 22 h with 78.96% formation rate (BR), field emission scanning electron microscopy (FESEM) results showed surface secreted EPS. Transcriptomic indicated associated stress response ( groS , mntH nth pdtaR pstA pstC radA rbpA whiB ybjG ), quorum sensing dppC livM luxS sapF polysaccharide metabolic process rfbX galE zwf opcA, glgC glgP gtfA ) may be In addition, 17 weighted gene co-expression network (WGCNA) modules identified two them positively correlated BR. Metabolomic amino acids amides; organic acids, alcohols esters; sugar (trehalose-6-phosphate, uridine diphosphategalactose, diphosphate-N-acetylglucosamine) main during These indicate response, (QS), EPS production are essential
