Abstract The composition and content of aroma substances in flue-cured tobacco ( Nicotiana tabacum L.) will affect the quality tobacco. To investigate correlation between various K326 before after flue-curing, their impact on diversity, this study employed middle leaves KRK26 as test materials. Samples were collected both flue-curing for untargeted metabolomics analysis. results showed that 584 metabolites significantly different there 44 aroma-related metabolites, including alcohols, aldehydes, phenols, organic acids, etc. analyzed compounds consist 37 known substances, while 7 previously not associated with aroma, have been identified other food products. These findings suggest these seven might may be potential compounds. Further analysis aldehydes increased but acids decreased. Furthermore, revealed a quantity type serve reference enhancing process optimizing industrial production cigarettes use cured leaves.

Introduction The main goal of tobacco fermentation technology is to minimize the alkaloid content while improving flavor substance content. Methods This study revealed microbial community structure and their metabolic functions during cigar leaf by high-throughput sequencing correlation analysis, evaluated performance functional microbes based on in vitro isolation bioaugmentation fermentation. Results relative abundance Staphylococcus Aspergillus increased first but then decreased fermentation, would occupy dominant position bacterial fungal communities, respectively, 21st day. Correlation analysis predicted that , Filobasidium could contribute formation saccharide compounds, Bacillus might have degradation effects nitrogenous substances. In particular, Candida as a co-occurring taxa biomarker later stage not only degrade substrates synthesize substances, also maintaining stability community. Moreover, inoculation, it was found parapsilosis metapsilosis significantly reduce alkaloids increase components leaves. Discussion validated critical role leaves through which help guide development starters directional regulation quality.

Metabolic enzyme activity and microbial composition of the air-curing fermentation processes determine quality cigar tobacco leaves (CTLs). In this study, we reveal evolution dominant microorganisms community structure at different stages CTLs. The results showed that changes in metabolic enzymes occurred mainly during phase, with polyphenol oxidase (PPO) being most active browning phase. Pseudomonas, Bacteroides, Vibrio, Monographella, Bipolaris, Aspergillus were key processes. Principal coordinate analysis revealed significant separation communities between phases. Redundancy bacteria such as Proteobacteria, Firmicutes, Bacteroidota, Acidobacteriota fungi Ascomycota Basidiomycota correlated temperature humidity. Bacteria act sugar metabolism, lipid amino acid while degrade lignin, cellulose, pectin through saprophytic action. Spearman correlation network Actinobacteria bacterial taxa, Dothideomycetes, Sordariomycetes, Eurotiomycetes fungal taxa. This research provides basis for improving cigars by KEY POINTS: • Changes POD PPO control color change CTLs stage. Aspergillus, Vibrio play an important role fermentation. Variations in industrial fermentation techniques have a significant impact on the of cigar tobacco leaves (CTLs), consequently influencing aromatic attributes resulting cigars. The entire process CTLs can be categorized into three distinct phases: phase 1 (CTLs prior to moisture regain), 2 post-moisture regain and pile fermentation), 3 after drying). These phases were determined based dynamic changes microbial community diversity. During 2, there was rapid increase total acid content, which facilitated proliferation Aerococcus, bacterial genus capable utilizing reducing sugars, malic acid, citric present leaves. In contrast, fungal microorganisms exhibited relatively stable response with Aspergillus, Alternaria, Cladosporium being dominant groups throughout stages. Bacterial genera found more closely associated variations volatile compounds during compared microorganisms. This association ultimately resulted higher levels aroma components CTLs, thereby improving overall quality findings reinforce significance shaping CTL provide valuable insights for future efforts artificial regulation secondary CTLs. KEY POINTS: • Industrial processes communities. Moisture content influence succession fermentation. strongly CTLs' aldehyde ketone flavors over fungi.

To elucidate the significant influence of microorganisms on geographically dependent flavor formation by analyzing microbial communities and volatile compounds (VFCs) in cigar tobacco leaves (CTLs) obtained from China, Dominica, Indonesia. Microbiome analysis revealed that predominant bacteria CTLs were Staphylococcus, Aerococcus, Pseudomonas, Lactobacillus, while fungi Aspergillus, Wallemia, Sampaiozyma. The different origins differed to some extent, diversity abundance greater than fungi. Metabolomic 64 VFCs identified, mainly ketones, which 23 could be utilized identify geographical CTLs. Sixteen with OAV 1, including cedrol, phenylacetaldehyde, damascone, beta-damascone, beta-ionone, play important roles shaping profile origins. Combined correlation analysis, bacterial more closely related key favored a positive correlation. Abstract This study delves into the aroma characteristics and microbial composition of filler tobacco leaves (FTLs) sourced from six distinct cigar-growing regions within Yunnan, China, following standardized fermentation. An integrated approach using gas chromatography-mass spectrometry (GC–MS), electronic nose (E-nose), microbiome analysis was employed for comprehensive profiling. Results derived Linear Discriminant Analysis (LDA) E-nose data confirmed presence notable variability in flavor substance profiles among FTLs regions. Additionally, GC–MS used to discern disparities volatile organic compound (VOC) distribution across these regions, identifying 92, 81, 79, 58, 69, 92 VOCs each respective sample set. Significantly, 24 emerged as pivotal determinants contributing heterogeneity diverse origins, indicated by Variable Importance Projection (VIP) analysis. Furthermore, distinctions free amino acid content chemical constituents were observed FTLs. Of noteworthy significance, solanone, isophorone, durene, (-)-alpha-terpineol, 2,3'-bipyridine exhibited strongest correlations with data, fungal microorganisms exerting a more pronounced influence on metabolites, elucidated through two-way orthogonal partial least-squares (O2PLS) modeling. These findings provide theoretical technical basis accurately evaluating synchronization aromas fermentation processes, they will enhance quality fermented foster growth domestic cigar industry ultimately.
