Among the most problematic bacteria with clinical relevance are carbapenem-resistant Enterobacterales (CRE), as there very limited options for their treatment. Treated wastewater can be a route release of these into environment and population. The aim this study was to isolate CRE from treated Zagreb treatment plant determine phenotypic genomic characteristics. A total 200 suspected were isolated, 148 which confirmed by MALDI-TOF MS. predominant species Klebsiella spp. (n = 47), followed Citrobacter 40) Enterobacter cloacae complex (cplx.) 35). All isolates carbapenemase producers multidrug-resistant phenotype. Using multi-locus sequence typing whole-genome sequencing (WGS), 18 different types identified among isolates, 14 associated human-associated clones. virulence gene analysis sequenced 7) revealed potential pathogenicity. PCR WGS showed that frequent genes in K. pneumoniae blaOXA-48 blaNDM-1, frequently occurred together, while blaKPC-2 together blaNDM-1 mainly detected oxytoca, E. cplx. Colistin resistance observed 40 % 57 isolates. Underlying mechanisms include known potentially novel intrinsic (point mutations pmrA/B, phoP/Q, mgrB crrB genes) acquired (mcr-4.3 gene). mcr-4.3 first time is probably located on conjugative IncHI1B plasmid. Considerable attention is given to intensive care unit-acquired infections; however, research on the transmission dynamics of multichain carbapenemase-resistant Enterobacter cloacae complex (CRECC) outbreaks remains elusive. A total 118 non-duplicated CRECC strains were isolated from clinical, intestinal, and hospital sewage samples collected Zhejiang province China during 2022–2023. 64 samples, their prevalence increased 10.0 % (95 confidence interval, CI = 0.52–45.8 %) in 2022 63.6 31.6–87.6 2023. Species-specific identification revealed that hormaechei was predominant species this study (53.4 %, 95 44.0–62.6 %). The antimicrobial susceptibility profiles indicated all conferred high-level resistance β-lactam antibiotics, ceftacillin/avibactam, polymyxin. The metagenomic surveillance of antimicrobial resistance in wastewater has been suggested as a methodological tool to characterize the distribution, status, and trends antibiotic-resistant bacteria. In this study, cross-sectional collection samples hospital-associated raw treated were obtained from February March 2020. Shotgun sequencing bioinformatic analysis performed bacterial abundance gene analysis. main phyla found all follows: Proteobacteria, Bacteroides, Firmicutes, Actinobacteria. At species level, ESKAPEE bacteria such E. coli relative decreased between wastewater, but S. aureus, A. baumannii, P. aeruginosa increased, did persistence K. pneumoniae both wastewater. A total 172 different ARGs detected; blaOXA, blaVEB, blaKPC, blaGES, mphE, mef, erm, msrE, AAC(6′), ant(3″), aadS, lnu, PBP-2, dfrA, vanA-G, tet, sul at highest persistence. Wastewater treatment plants are hotspots for the release of antimicrobial resistant pathogenic bacteria into aquatic ecosystems, significantly contributing to cycle resistance. Special attention should be paid ESKAPE bacteria, which have been identified as high-priority targets control measures. Among them, Klebsiella pneumoniae is particularly noteworthy. In this study, we collected wastewater samples from inlet, sedimentation tank, and effluent water a plant in June, July, October, November 2018. We detected characterized 42 K. strains using whole genome sequencing (15 8 19 effluent). Additionally, were tested their resistance phenotype. Using no distinct patterns observed terms genetic profiles. All tetracycline, meanwhile 60%, 47%, 37.5% isolated effluent, respectively, multidrug resistant. Some isolates also colistin, nearly all positive eptB arnT genes, associated with polymyxin Various genes linked mobile elements, they did not correlate virulence groups or defense systems. Overall, our results, although quantitative, highlight that strains, including those colistin genetically unrelated, being discharged ecosystems plants. This suggests necessity monitoring aimed at characterizing these bacteria.

It has been debated whether wastewater treatment plants (WWTPs) primarily act to attenuate or amplify antibiotic resistance genes (ARGs). However, ARGs are highly diverse with respect their mechanisms, mobilities, and taxonomic hosts therefore behavior in WWTPs should not be expected universally conserved. We applied metagenomic sequencing influent effluent samples from 12 international classify the of specific entering exiting WWTPs. In total, 1079 different originating a variety bacteria were detected. This included that could mapped assembled scaffolds corresponding nine human pathogens. While relative abundance (per 16S rRNA gene) decreased during at 11 sampled absolute mL) all WWTPs, increases observed for 40% detected 12th WWTP.
