This study aimed to characterize virulence and antibiotic resistance genes in multidrug-resistant (MDR) extensively drug-resistant (XDR) Escherichia coli Klebsiella pneumoniae isolated from cases of subclinical mastitis (SCM) buffaloes. A cross-sectional was conducted on 1540 quarter milk samples collected 385 Milk were screened using the California Mastitis Test Modified Whiteside Test. Positive underwent bacterial culture, biochemical tests, biofilm detection molecular analysis for pathogen identification virulence, resistance, extended-spectrum beta-lactamase (ESBL) genes. The prevalence SCM 67.9% (1046/1540) at level 80.8% (311/385) animal level. E. identified 9.5% (146/1540) samples, while K. detected 9.09% (140/1540). Virulence genes, such as stx1 (27.4%), including aac(3)-iv (77.4%) tetA (76.7%), exhibited higher prevalence. Additionally, β-lactamase notably blaTEM (67.1%), ESBL blaCTX-M1, detected. Biofilm formation 83.6% (122/146) isolates 75.7% (106/140) isolates. Antimicrobial susceptibility testing revealed significant ampicillin, amoxicillin-clavulanic acid, aminoglycosides. MDR observed 31.5% 39.3% isolates, with XDR rates 8.9% 12.9%, respectively. These findings underscore alarming spread resistant pathogens SCM-affected buffaloes, emphasizing urgent need ongoing surveillance targeted intervention strategies.
