We investigated a mass mortality event of Mediterranean mussels (Mytilus galloprovincialis) in the Novigrad Sea, Croatia during winter 2022/2023. The rate was up to 70% and initially linked an influx freshwater from Zrmanja River. Despite mitigation measures that included placing deeper avoid influence freshwater, mortalities continued. Subsequent analysis 64 samples using cytology, histolology real-time PCR exclude listed diseases, yielded negative results. testing for Haplosporidium pinnae revealed presence haplosporidian-like DNA resembling Minchinia mytili. In study predominant bacteria, Psychrobacter sp., Colwellia Vibrio splendidus were detected. The New Zealand Greenshell™ mussel (Perna canaliculus) is an economically important aquaculture species. Prolonged increases in seawater temperature above thermotolerance ranges pose a significant threat to survival and health, potentially increasing susceptibility bacterial infections. Using challenge experiments, this study examined the combined effects of increased (Photobacterium swingsii) infection on animal survival, haemocyte biochemical responses adult mussels. Mussels maintained at three temperatures (16, 20 24 °C) for seven days were either not injected (control), with sterile marine broth (injection control) or P. swingsii (challenged medium high doses) monitored daily five days. Haemolymph tissue samples collected 24, 48, 72, 96, 120 h post-challenge analysed quantify colonies, responses. infected exhibited mortalities °C, likely due compromised immune system, but no observed when was only stressor. Bacterial colony counts haemolymph decreased over time, suggesting clearance followed by activation signalling pathways. Total viability data supports defence functions being stimulated presence pathogen loads °C. In gill tissue, oxidative stress responses, measured as total antioxidant capacity malondialdehyde (MDA) levels, higher mussels (compared controls) after 24h 120h lowest (16 highest (24 °C), indicating stressors. Overall, work confirms that Photobacterium pathogenic canaliculus indicates may be more vulnerable pathogens under conditions elevated temperature, such those predicted future climate change scenarios.

However, the underlying mechanisms at cellular level are not completely understood. Metabolomics has become valuable tool to evaluate responses marine organisms various stressors. Broadcast spawners, like the blue mussel Mytilus edulis, experience substantial energy expenditure during spawning due to extensive gamete release that can divert from other functions. This energetic cost might be intensified by environmental stressors, including hypoxia suppress aerobic metabolism. However, implications of in marine broadcast spawners have not been well studied. We examined effects short-term (7 days) and on mitochondrial activity, reactive oxygen species (ROS) production, cellular allocation (ratio tissue reserves demand) somatic tissues M. edulis. Under normoxic conditions, post-spawning (72 h) recovery correlated with increased phosphorylation (OXPHOS) rate mitochondria digestive gland, while inhibited this response. Regardless levels, ROS production decreased after spawning, indicating edulis' ability prevent oxidative stress. Spawning led reduced (the gills gland), highlighting significant primarily fueled lipid protein breakdown. Additionally, dropped 3 h post-spawning, a shift demand supply. Normoxic conditions allowed 72 h, but hindered recuperation. These findings underscore spawning's bioenergetic challenge for potentially elevating mortality risk, especially hypoxic coastal habitats.

Abstract For more than a century, bacterial infections caused by Vibrio spp. have affected various species of mussels around the world, with limited insights into responses implemented against these infections. A combination chemical analyses and carefully selected biological endpoints from haemolymph tissues can be used to identify welfare status potentially protect aquatic ecosystems catastrophic health threats. Recent developments in biomarker identification tools, such as omics bioinformatics, been successfully applied evaluate effect environmental pollutants other chemicals on mussels. However, application biomarkers assess mussel is limited. This review describes available scientific literature research for -mussel interactions, those aspects related disease assessment, grouped exposure, effects, susceptibility. From review, it clear that when integrated are used, they provide deeper understanding relative potential susceptibility better management practices. Furthermore, data build resilience climate change conditions, strengthen biosecurity programs, improve farming processing efficiency, add value terms market-desirable traits. Gender differences in the hemocyte immune response of Hong Kong oyster Crassostrea hongkongensis to Vibrio harveyi and lipopolysaccharide (LPS) infection exist. To determine if a gender difference also exists, we use 1H NMR-based metabolomics method investigate responses C. hepatopancreas tissues V. LPS infection. Both infections induced pronounced gender- immune-specific metabolic tissues. Responses are mainly presented changes substances involved energy metabolism (decreased glucose, ATP, AMP males increased ATP LPS-infected females), oxidative stress glutathione decreased tryptophan phenylalanine choline proline tricarboxylic acid (TCA) cycle α-ketoglutarate fumarate males, osmotic regulation trigonelline taurine harveyi-infected betaine females). Results suggest that post-spawning-phase male oysters have more significant greater ability cope with than female oysters.
