Autism spectrum disorder (ASD) is a behaviorally defined neurodevelopmental characterized by deficits in language, communication, and social function with an estimated prevalence rate of between 1 30 44 U.S. births. Gene/environment (G × E) interactions are widely regarded as the most probable explanation for idiopathic ASD, especially because some genes selectively targeted various environmental xenobiotics. Because deciduous teeth likely biomarker utero exposure, present study investigated if quantity chemicals found differs children without ASD. Twenty-two from ASD 20 typically developed were prepared analyzed using THE Two-Dimensional Gas Chromatography Time-of-Flight Mass Spectrometer (GC GC-TOF MS) ChromaTOF version 23H2 software Agilent 7890 gas chromatograph. The autism sample had significantly more their than typical developing (99.4 vs. 80.7, respectively) (p < 0.0001). majority identified phthalates, plasticizers, pesticides, preservatives, or intermediary solvents used production fragranced personal care cleaning products flavoring agents foods. known toxic analytes reported this biomarkers developmental exposure. Why there greater concentrations that came unclear. A further understanding cavalcade multiple biological system (Interactome) could help future efforts to reduce risks. Introduction Critical phases of neurodevelopment and gut microbiota diversification occur in early life both processes are impacted by genetic environmental factors. Recent studies have shown the presence alterations neurodevelopmental disorders. Here we performed a systematic review intestinal composition function pediatric adult patients affected autism spectrum disorder (ASD), attention-deficit/hyperactivity (ADHD), Rett syndrome (RETT). Methods We searched selected keywords online databases PubMed, Cochrane, OVID (January 1980 to December 2021) with secondary references eligible articles. Two reviewers independently critical appraisals on included articles using Appraisal Skills Program for each study design. Results Our identified 18, 7, 3 original describing profiles ASD, ADHD, RETT, respectively. Decreased Firmicutes increased Bacteroidetes were observed individuals ASD ADHD. Proinflammatory cytokines, short-chain fatty acids neurotransmitter levels altered RETT. Constipation visceral pain related changes Hyperactivity impulsivity negatively correlated Faecalibacterium (phylum Firmicutes) positively Bacteroides sp. Bacteroidetes) ADHD subjects. Five explored microbiota-or diet-targeted interventions Probiotic treatments Lactobacillus fecal transplantation from healthy donors reduced constipation ameliorated symptoms children. Perinatal administration prevented onset Asperger adolescence. Micronutrient supplementation improved disease symptomatology without causing significant communities’ composition. Discussion Several discrepancies found among studies, primarily due sample size, variations dietary practices, high prevalence functional gastrointestinal symptoms. Abstract The etiologic pathways leading to neuropsychiatric diseases remain poorly defined. As genomic technologies have advanced over the past several decades, considerable progress has been made linking disorders genetic underpinnings. Interest and consideration of nongenetic risk factors (e.g., lead exposure schizophrenia) have, in contrast, lagged behind heritable frameworks explanation. Thus, association illness environmental chemical exposure, their potential interactions with susceptibility, are largely unexplored. In this review, we describe emerging approaches for considering impact acting alone concert risk, point role epigenetics mediating effects on transcription genes implicated mental disorders. We highlight recent examples research psychiatric that shared biological mechanisms—synaptic dysfunction, immune alterations, gut–brain interactions. outline new tools resources can be harnessed study These tools, combined experimental evidence, suggest there is a need broadly incorporate exposures research, ultimate goal identifying modifiable informing treatment strategies disease.

Autism spectrum disorder (ASD) and Alzheimer’s disease (AD) are neurodevelopmental neurodegenerative disorders affecting two opposite ends of life span, i.e., childhood old age. Both pose a cumulative threat to human health, with the rate incidences increasing considerably worldwide. In context recent developments, we aimed review correlated symptoms genetics, overlapping aspects in mechanisms pathogenesis ASD AD. Dementia, insomnia, weak neuromuscular interaction, as well communicative cognitive impairments, shared symptoms. A number genes proteins linked both have been tabulated, including MECP2, ADNP, SCN2A, NLGN, SHANK, PTEN, RELN, FMR1. Theories about role neuron development, processing, connectivity, levels neurotransmitters discussed. Based on literature, roles FMRP (Fragile X mental retardation protein), hnRNPC (heterogeneous ribonucleoprotein-C), IRP (Iron regulatory proteins), miRNAs (MicroRNAs), α-, β0, γ-secretases posttranscriptional regulation cellular synthesis processing APP (amyloid-β precursor protein) elaborated describe parallel routes AD pathogenesis. However, interactive genetic environmental factors, oxidative metal ion stress, mutations associated genes, alterations related pathways development needs further investigation.
