Understanding neuromodulatory effects of serotonin 2A receptor (5-HT R) agonists with diverse pharmacological profiles is relevant to advancing psychedelic-related drug applications. We performed simultaneous positron emission tomography (PET) and magnetic resonance imaging (phMRI) in anesthetized nonhuman primates (NHP; N = 3) examine partial varying 5-HT R affinities selectivity profiles: psilocybin (30, 60, 90 µg/kg), lisuride (5 25CN-NBOH (15 µg/kg). Background: Psychedelics are increasingly recognized as a promising and innovative treatment strategy for several mental disorders. However, there is still lively controversy in the medical community regards rationale of their employment, specifically indications potential dangers. Methods: A comprehensive literature search on “MEDLINE/PubMed” “Web Science” was performed from inception to 26 June 2024, cross-checking obtained references. We included all studies, i.e., both clinical preclinical, that supplied original data. Results: initially total 1083 entries, 813 MEDLINE/PubMed 270 Web Science. After duplicate elimination, 903 underwent systematic selection. Primary abstract screening yielded 572 candidates eligibility assessment excluded 331 entries formal grounds. Psilocybin, a naturally occurring psychedelic compound, has garnered renewed scientific interest for its potential in treating psychiatric and neurological disorders. This review systematically examines the latest research on psilocybin’s pharmacokinetics, pharmacodynamics, clinical efficacy, safety profile. Emerging evidence supports efficacy conditions such as major depressive disorder (MDD), treatment-resistant depression (TRD), anxiety, alcohol use disorders (AUD), cancer-related distress. Despite promising outcomes, significant barriers remain, including methodological constraints, regulatory hurdles, limited population diversity trials. Psilocybin is a hallucinogen with complex neurobiological and behavioral effects. This the first study to use MRI follow functional changes in brain activity response different doses of psilocybin fully awake, drug naive rats. We hypothesized that would show dose-dependent increase prefrontal cortex thalamus, while decreasing hippocampal activity. Female male rats were given IP injections vehicle or 0.03 mg/kg, 0.3 3.0 mg/kg awake during imaging session. These levels validated by measuring its metabolite, psilocin. Changes BOLD signal recorded over 20 min window. Data for resting state connectivity collected approximately 35 post injection. All data registered rat 3D atlas 169 areas providing site-specific global connectivity. Treatment resulted significant positive signal. The most affected acute presentation somatosensory cortex, basal ganglia thalamus. Males females showed sensitivity dose, exhibiting greater activation than males at especially thalamic regions. Abstract The use of psilocybin to treat alcohol disorder is very promising, but its mechanisms action remain poorly understood. We combined behavioural, pharmacological and gene expression analyses decipher the psilocybin, for first time, when injected into brain. Male Long Evans rats underwent chronic operant ethanol self-administration before testing effect intraperitoneal or directly within nucleus accumbens core ventral tegmental area. Transcripts from dopaminergic system were quantified in prefrontal cortex. Psilocybin significantly reduced (by 50%) 4 h session either intraperitoneally (1 mg/kg) left (0.15 μg) not right injection was prevented by intra-left 0.3 μg 5-HT2A receptor antagonist ketanserin. In that self-administered those self-administering saccharin, dopamine D2 (D2R) mRNA increased both cortex while D1 only As humans, through accumbens, possibly D2R expression. Background: Recent evidence suggests that psychedelics are able to induce rapid and long-lasting antidepressant effects. The generally acknowledged explanation for these traits is the phenomenon of neuroplasticity, although exact underlying molecular mechanisms remain unclear. Aims: This study investigates selected neuroplastic effects psilocin, lysergic acid diethylamide (LSD) N,N-dimethyltryptamine (DMT) in direct comparison with ketamine, fluoxetine lithium after acute (1 h) and/or prolonged (24 treatment vitro. Methods: Rat primary cortical cultures were treated 10 uM 1 (LSD), 90 N, N-dimethyltryptamine (DMT), 5 mM lithium.
