Abstract Neurological conditions, including cognitive impairment and Alzheimer’s disease (AD), impose a huge burden on society, affecting millions of people globally. In addition to genetic factors, recent studies indicate that environmental experiential factors may contribute the pathogenesis these diseases. Early life adversity (ELA) has profound impact brain function health later in life. rodent models, exposure ELA results specific deficits aggravated AD pathology. Extensive concerns have been raised regarding higher risk developing impairments with history ELA. this review, we scrutinize findings from human animal focusing connection AD. These discoveries suggest ELA, especially at early postnatal stages, increases susceptibility terms mechanisms, could lead dysregulation hypothalamus-pituitary-adrenal axis, altered gut microbiome, persistent inflammation, oligodendrocyte dysfunction, hypomyelination, aberrant adult hippocampal neurogenesis. Abstract Childhood maltreatment has been suggested to have an adverse impact on neurodevelopment, including microstructural brain abnormalities. Existing neuroimaging findings remain inconsistent and heterogeneous. We aim explore the most prominent robust cortical thickness (CTh) gray matter volume (GMV) alterations associated with childhood maltreatment. A systematic search relevant studies was conducted through September 2022. The whole-brain coordinate-based meta-analysis (CBMA) CTh GMV were using seed-based d mapping (SDM) software. Meta-regression analysis subsequently applied investigate potential associations between clinical variables structural changes. total of 45 eligible for inclusion, 11 datasets 39 GMV, consisting 2550 participants exposed 3739 unexposed comparison subjects. Individuals exhibited overlapped deficits in median cingulate/paracingulate gyri simultaneously revealed by both GM studies. Regional thinning right anterior left middle frontal gyrus, as well reductions supplementary motor area (SMA) also identified. No greater regions found either or GMV. In addition, several neural morphology changes average age maltreated individuals. might serve feature effects early-life trauma human predominantly involved cognitive functions, socio-affective functioning stress regulation. Abstract While a myriad of factors likely contribute to the development mental illness in young people, social environment (including early adverse experiences) concert with neurodevelopmental alterations is undeniably important. A number influential theories make predictions about how and why may mediate or moderate effects on emergence illness. Here, we discuss current evidence supporting each these theories. Although this area research rapidly growing, body still relatively limited. However, there exist some consistent findings, including increased striatal reactivity during positive affective processing larger hippocampal volumes being associated vulnerability susceptibility environments internalizing symptoms. Limited longitudinal work has investigated mechanisms linking health. Drawing from human insights animal studies, propose an integrated mediation-moderation model outline future directions advance field.

Abstract Complementary transistors are critical for circuits with compatible input/output signal dynamic range and polarity. Organic electronics offer biocompatibility conformability; however, generation of complementary organic requires introduction separate materials inadequate stability potential tissue toxicity, limiting their use in biomedical applications. Here, we discovered that source/drain contact asymmetry enables spatial control de/doping creation single-material from a variety conducting polymers both carrier types. When integrated the vertical channel design internal ion reservoirs ion-gated electrochemical transistors, produced matched IGTs (cIGTs) formed high-performance conformable amplifiers 200 V/V uniform gain 2 MHz bandwidth. These showed long-term vivo stability, miniaturized biocompatible allowed implantation developing rodents to monitor network maturation. cIGTs expand standard circuit designs enhance potential.

Mother-infant interactions during the early postnatal period are critical for infant survival and scaffolding of development. Rodent models used extensively to understand how these social experiences influence neurobiology across lifespan. However, methods measuring dam-pup typically involve time-consuming manual scoring, vary widely between research groups, produce low density data that limits downstream analytical applications. To address methodological issues, we developed Automated Maternal Behavior Early life in Rodents (AMBER) pipeline quantifying home-cage maternal mother-pup using open-source machine learning tools. DeepLabCut was track key points on rat dams (32 points) individual pups (9 per pup) day 1-10 video recordings. Pose estimation reached point test errors approximately 4.1-10 mm (14.39 pixels) 3.44-7.87 (11.81 depending depth animal frame averaged all dam respectively. human-annotated behavior labels from 38 videos were with Simple Behavioral Analysis (SimBA) generate classifiers active nursing, passive nest attendance, licking grooming, self-directed eating, drinking random forest algorithms. A set 242 AMBER produced measures expected range videos. Abstract Early-life stress (ELS) leaves signatures upon the brain that persist throughout lifespan and increase risk of psychiatric illnesses including mood anxiety disorders. In humans, myriad forms ELS—including childhood abuse, bullying, poverty, trauma—are increasingly prevalent. Understanding signs ELS, those associated with illness, will enable improved treatment prevention. Here, we developed a novel procedure to model human ELS in mice identify translationally-relevant biomarkers We exposed male (C57BL/6 J) an early-life (juvenile) chronic social defeat (jCSDS) examined interaction responsivity reward during adulthood. As expected, jCSDS-exposed showed socially avoidant phenotype open-field tests. However, sucrose preference tests failed demonstrate ELS-induced reductions choice for sweetened solution, suggesting no effect on function. To explore whether other tasks might be more sensitive changes motivation, tested Probabilistic Reward Task (PRT), often used humans study learning deficits depressive illness. touchscreen PRT variant was reverse-translated maximize alignment version subjects, jCSDS displayed significant tendency develop response biases richly-rewarded stimulus, hallmark sign anhedonia when observed humans. Our findings suggest procedures utilize same endpoints across species may development systems accurately predict outcomes

Early-life adversity (ELA) increases the likelihood of neuropsychiatric diagnoses, which are more prevalent in women than men. Since changes reproductive hormone levels can also increase probability anxiety disorders women, we examined effects ELA on adult female mice across estrous cycle. We found that during diestrus, when progesterone relatively high, exhibit increased avoidance behavior and theta oscillation power ventral hippocampus (vHIP). diestrus had higher lower allopregnanolone, a neurosteroid metabolite progesterone, vHIP compared with control-reared mice. Progesterone receptor antagonism normalized mice, while treatment negative allosteric modulator allopregnanolone promoted control These results suggest altered signaling
