Issues caused by maxillofacial tumours involve not only dealing with but also repairing jaw bone defects. In traditional tumour therapy, the systemic toxicity of chemotherapeutic drugs, invasive surgical resection, intractable recurrence, and metastasis are major threats to patients' lives in clinic. Fortunately, biomaterial-based intervention can improve efficiency treatment decrease possibility recurrence metastasis, suggesting new promising antitumour therapies. addition, tissue defects their negatively affect physiological psychological health patients, investment result a multitude burdens society. Biomaterials options because they have good biocompatibility bioactive properties for stimulation regeneration. More interestingly, an integrated material regimen that combines therapy repair is option. Herein, we summarized biomaterial-mediated treatments analysed biomaterials defect repair. Furthermore, proposed superior design dual-functional simultaneous regeneration provide strategy managing quality life patients future.

Oral cancers constitute the majority of head and neck tumors, with a relatively high incidence poor survival rate in developing countries. While five-year rates oral cancer patients have increased to 65%, overall for advanced stages has been at 27% past ten years, emphasizing necessity further understanding etiology disease, diagnosis, formulating possible novel treatment regimens. MicroRNAs (miRNAs), family small non-coding RNA, emerged as master modulators gene expression various cellular biological process. Aberrant these dynamic molecules associated many human diseases, including cancers. The deregulated miRNAs shown control oncogenic processes, sustaining proliferative signaling, evading growth suppressors, resisting cell death activating invasion metastasis, inducing angiogenesis. Hence, aberrant cancers, makes them potential candidates investigation functional markers, which will aid differential prognosis, development therapeutic This review presents holistic insight into our role regulating hallmarks tumorigenesis.

Oral squamous cell carcinoma (OSCC) is the most common malignancy of oral cavity and accounts for >90% all cancers. Despite advances in diagnostic procedures therapeutic interventions, overall survival has not improved significantly recent decades, primarily due to late diagnosis, locoregional recurrence treatment resistance. Identifying reliable biomarkers early detection, prognosis evaluation response prediction critical improving clinical outcomes patients with OSCC. In present review, prognostic predictive utility circulating biomarkers, such as tumour cells, serological histological genetic were explored context addition, potential role immune checkpoints OSCC was highlighted rapidly evolving field liquid biopsy its revolutionize examined. The existing evidence these critically evaluated challenges limitations associated their introduction into routine practice addressed. conclusion, review highlights promising current understanding pathogenesis offers avenues patient care through personalized medicine approaches.

The association between pretreatment nutritional status and immunotherapy response in patients with advanced head neck cancer is unclear. We retrospectively analyzed a cohort of 99 who underwent treatment anti-PD-1 or anti-CTLA-4 antibodies (or both) for stage IV HNSCC 2014 2020 at the Johns Hopkins Hospital. Patient demographics clinical characteristics were retrieved from electronic medical records. Baseline prognostic index (PNI) scores body mass (BMI) trends calculated. Associations PNI BMI correlated overall survival (OS), progression-free (PFS), response. In univariate analysis, there was significant correlation OS PFS baseline (OS: HR: 0.464; 95% CI: 0.265-0.814; PFS: p = 0.007 0.525; 0.341-0.808; 0.003). Poor also associated greater decrease trend (HR: 0.42; 0.229-0.77; 0.005). multivariate but not significantly log (HR) -0.79, -1.6, -0.03, 0.041; -0.78, -1.4, -0.18, 0.011). conclusion, poor negative post-immunotherapy outcomes.

Background: The programmed cell death ligand 1/programmed receptor 1 (PD-L1/PD-1) Immune Checkpoint is an important modulator of the immune response. Overexpression and its ligands involved in immunosuppression failure response against tumor cells. PD-1/PD-L1 overexpression oral squamous carcinoma (OSCC) compared to healthy mucosa (NOM) has already been demonstrated. However, little known about expression precancerous lesions like leukoplakia (OLP). aim study was investigate whether increased exists OLP it associated with malignant transformation. Material Methods: PD-1 PD-L1 immunohistologically analyzed separately epithelium (E) subepithelium (S) that had undergone transformation within 5 years (T-OLP), without (N-OLP), corresponding OSCC NOM. Additionally, RT-qPCR analysis for done entire tissues. association between transformation, dysplasia inflammation were examined. Results: Compared N-OLP, there levels protein epithelial subepithelial layers T-OLP (pE = 0.001; pS 0.005). There no significant difference mRNA N-OLP (p 0.128), but fold-change increase these groups (Relative Quantification (RQ) 3.1). In contrast significantly 0.007), not 0.25). Importantly, years. Conclusion: Increased are related may represent a promising prognostic indicator determine risk progression OLP. might establish immunosuppressive microenvironment, which could favor escape thereby contribute Hence, checkpoint inhibitors counteract development serve as efficient therapeutic strategy patients high-risk lesions.

Head and neck cancer (HNC), also known as the that can affect structures between dura mater pleura, is 6th most common type of cancer. This heterogeneous group malignancies usually treated with a combination surgery radio- chemotherapy, depending on if disease localized or at an advanced stage. However, HNC patients are diagnosed stage, resulting in death half these patients. Thus, prognosis recurrent/metastatic HNC, especially squamous cell carcinoma (HNSCC), notably poorer than HNC. review explores epidemiology etiologic factors histopathology this cancer, diagnosis methods treatment approaches currently available. Moreover, special interest given to novel therapies used treat subtypes worse prognosis, exploring immunotherapies targeted/multi-targeted drugs undergoing clinical trials, well light-based (i.e., photodynamic photothermal therapies).
