Ferroptosis is a type of regulated cell death characterized by iron-mediated lipid peroxidation, in contrast with apoptosis, autophagy, and necrosis. It can be triggered many pathological processes, including cellular metabolism, tumors, neurodegenerative diseases, cardiovascular ischemia-reperfusion injuries. In recent years, ferroptosis has been discovered to associated p53. P53 tumor suppressor protein multiple powerful functions cycle arrest, senescence, death, repair DNA damage, mitophagy. Emerging evidence shows that plays crucial role suppression as key bidirectional regulator adjusting metabolism iron, lipids, glutathione peroxidase 4, reactive oxygen species, amino acids via canonical pathway. addition, noncanonical pathway p53 regulates years. The specific details require further clarified. These mechanisms provide new ideas for clinical applications, translational studies have performed treat various diseases.

ALK-positive NSCLC patients demonstrate initial responses to ALK tyrosine kinase inhibitor (TKI) treatments, but eventually develop resistance, causing rapid tumor relapse and poor survival rates. Growing evidence suggests that the combination of drug immune therapies greatly improves patient survival; however, due low immunogenicity tumors, do not respond currently available immunotherapies. Tumor-associated macrophages (TAMs) play a crucial role in facilitating lung cancer growth by suppressing tumoricidal activation absorbing chemotherapeutics. However, they can also be programmed toward pro-inflammatory suppressive phenotype, which represents highly active area therapy development. Iron loading TAMs achieve such reprogramming correlating with an improved prognosis patients. We previously showed superparamagnetic iron oxide nanoparticles containing core-cross-linked polymer micelles (SPION-CCPMs) target stimulate activation. Here, we show SPION-CCPMs secrete reactive nitrogen species cytokines exert activity. further reshape immunosuppressive

A new type of nonapoptotic, iron-dependent cell death induced by lipid peroxidation is known as ferroptosis. Numerous pathological processes, including inflammation and cancer, have been demonstrated to be influenced changes in the ferroptosis-regulating network. Long non-coding RNAs (LncRNAs) are a group functional RNA molecules that not translated into proteins, which can regulate gene expression various manners. An increasing number studies shown lncRNAs interfere with progression ferroptosis modulating ferroptosis-related genes directly or indirectly. Despite evidence implicating cancer inflammation, on their mechanisms therapeutic potential remain scarce. We investigate lncRNA-mediated regulation immunity, assessing feasibility challenges targets these conditions.

As a novel form of regulated cell death, ferroptosis is characterized by intracellular iron and lipid peroxide accumulation, which different from other death forms morphologically, biochemically, immunologically. Ferroptosis metabolism, antioxidant defense systems as well various transcription factors related signal pathways. Emerging evidence has highlighted that associated with many physiological pathological processes, including cancer, neurodegeneration diseases, cardiovascular ischemia/reperfusion injury. Noncoding RNAs are group functional RNA molecules not translated into proteins, can regulate gene expression in manners. An increasing number studies have shown noncoding RNAs, especially miRNAs, lncRNAs, circRNAs, interfere the progression modulating ferroptosis-related genes or proteins directly indirectly. In this review, we summarize basic mechanisms regulations focus on recent mechanism for types ncRNAs to conditions, will deepen our understanding regulation provide new insights employing ferroptosis-associated therapeutic strategies.

Gastric Cancer (GC) is a prevalent malignancy within the digestive tract, ranking as fifth most common malignant tumor worldwide. It characterized by clinical features such tendency for metastasis and an unfavorable prognosis. Ferroptosis, recently identified form of cell death, represents novel mode cellular demise that diverges from traditional concepts necrosis apoptosis. Numerous studies have found ferroptosis plays significant role in proliferation, metastasis, drug resistance, microenvironment regulation GC. This review summarizes mechanism its occurrence development GC cells. provides examples demonstrating how various anti-tumor drugs can induce Additionally, it potential application value future treatment
