Objective: Mendelian randomization (MR) has been widely utilized for causal inference between diseases, and its implementation within the domain of traditional Chinese medicine (TCM) is considered feasible. Although previous clinical epidemiological studies have demonstrated a close relationship insomnia depression, inherent genetic factors underlying these associations are unclear. The aim this study was to evaluate depression via bidirectional 2-sample MR increase understanding TCM theory treating different diseases with same method, particularly in context comorbid insomnia. Methods: Genetic data related were extracted from published genome-wide association (GWAS) sets. Single-nucleotide polymorphisms (SNPs) associated used as instrumental variables construct an “SNP-exposure-outcome” model. Bidirectional analysis conducted inverse-variance weighted (IVW), median, Egger regression, simple mode, mode methods. Furthermore, heterogeneity tests, pleiotropy analyses, sensitivity analyses performed. Results: results revealed increased risk developing (IVW, OR=1.400, 95% CI: 1.246–1.573, P <0.001), OR=1.204, 1.144–1.266, <0.001). As a traditional Chinese medicine formula used in clinical practice for an extended period, Suxin-Hugan-Fang (SXHGF) exhibits excellent anti-inflammatory properties. However, the efficacy of SXHGF treating ulcerative colitis (UC) and its mechanism action are still unclear. In this study, therapeutic effects on UC were evaluated using network pharmacology experimental validation, while also investigating action. By administering DSS to C57BL/6 mice construct mouse model colitis, effect was based weight loss percentage, disease activity index, colon length changes, pathological conditions as indicators. The main chemical components determined by LC-MS-QTOF method. potential targets mechanisms treatment inferred bioinformatics methods, further validated through ELISA, IHC, Western blotting assays. results demonstrate that can suppress oxidative stress damage tissue mice, alleviate DSS-induced inhibiting JAK2/STAT3 NFκB pathways.
