Abstract Grapevine is an important economic fruit tree worldwide, but grape production has been plagued by a vast number of fungal diseases, which affect vigor and the quality yield berries. To seek remedies for such issues, researchers have always committed to conventional biotechnological breeding. In recent years, increasing progress made in elucidating molecular mechanisms grape–pathogenic fungi interactions resistance regulation. This paper focuses on the creation of an in vitro collection grapevine hybrids from breeding program Kazakh Scientific Research Institute Fruit Growing and Viticulture investigates presence Plasmopara viticola resistance mediated by Rpv3 Rpv12 loci. We looked at optimization establishment using either shoots taken directly field-grown plants or budwood cuttings forced indoors. further screened for endophyte contamination initiated explants optimized multiplication stage. Finally, loci against P. was studied. The field-sourced were more effective method providing plant sources initiation once all accessions met goal establishment. concentration phytohormones acidity culture medium have a great effect rate quality stock cultures. Out 17 accessions, 16 showed single combined viticola. identified as carrying alleles represent important genetic resources disease programs. Vincetoxicum spirale (Forssk.) D.Z.Li, a threatened perennial climber with therapeutic values, belongs to the family Apocynaceae (Subfamily: Asclepiadoideae). Constraints related seeds and seedling establishment, coupled wide-spread habitat destruction depleted natural populations of this species. This study presents first report an efficient indirect organogenesis protocol using mature leaf-derived callus V. spirale. Of four types explants evaluated (leaf, node, internode root), leaf were found be most suitable for culture establishment de novo organogenesis. Leaf cultured on Murashige Skoog (MS, 1962) medium containing 2.0 mg L−1 2,4-dichlorophenoxyacetic acid (2,4-D) showed highest response (76.7%) induction. Optimum proliferation was achieved MS supplemented 1.0 2,4-D 0.5 Kinetin (Kin), which produced morphogenic, competent, light-green-friable callus. Abstract Key message Gene silencing of BcDCL genes improves gray mold disease control in the cultivated strawberry. technology offers new opportunities to develop formulations or pathogen-resistant plants for reducing impacts agricultural systems. Recent studies offered proof concept that symptoms can be reduced by downregulating Dicer-like 1 ( DCL1 ) and 2 DCL2 Botrytis cinerea . In this study, we demonstrate both solutions based on dsRNA topical treatment planta expression targeting BcDCL1 BcDCL2 used strawberry mold, most harmful different fruit crops. 50, 70 100 ng μL −1 naked BcDCL1/2 dsRNA, sprayed Fragaria x ananassa cultivar Romina greenhouse, displayed significant reduction susceptibility, compared negative controls, but a lesser extent than chemical fungicide.
