Background: Subthreshold depression (StD) is considered to be the "precursor" stage of major depressive disorder (MDD), which could cause higher risk suicide, disease burden and functional impairment. There have been various non-pharmacological interventions for StD. However, comparison their effectiveness still lacks sufficient evidence. We performed a systematic review network meta-analysis evaluate rank efficacy multiple targeting Methods: conducted thorough search across databases including PubMed, Medline, Embase, Web Science PsycINFO from inception December 2022. All included studies were randomized controlled trials (RCTs) patients with StD compared control group (CG). Several universal scales measuring severity used as outcomes. The surface under cumulative ranking curve (SUCRA) was separately each intervention using "Stata 17.0" software. Results: A total thirty-six included, involving twenty-eight 7417 participants. research found that most superior controls In outcome evaluation by different depression, psychotherapy always ranked first in terms treatment effectiveness, especially Problem-solving Therapy (PST), Behavioral Activation (BAT), Cognitive (CBT)/Internet-based CBT (I-CBT)/Telephone-based (T-CBT). Migraine is a neurovascular disorder that can be debilitating for individuals and society. Current research focuses on finding effective analgesics management strategies migraines by targeting specific receptors neuropeptides. Nonetheless, newly approved calcitonin gene-related peptide (CGRP) monoclonal antibodies (mAbs) have 50% responder rate ranging from 27 to 71.0%, whereas CGRP receptor inhibitors 56 71%. To address the need novel therapeutic targets, researchers are exploring potential of another secretin family peptide, pituitary adenylate cyclase-activating polypeptide (PACAP), as ground-breaking treatment avenue migraine. Preclinical models revealed how PACAP affects trigeminal system, which implicated in headache disorders. Clinical studies demonstrated significance migraine pathophysiology; however, few clinical trials remain inconclusive: 1 mAb, AMG 301 showed no benefit prevention, while ligand Lu AG09222 significantly reduced number monthly days over placebo phase 2 trial. Meanwhile, vasoactive intestinal (VIP) gaining interest new target. In light recent advances research, we emphasize promising target treatment, highlighting member antimigraine armamentarium, especially patients who do not respond or contraindicated anti-CGRP therapies. By updating our knowledge its unique contribution pathophysiology, pave way reinforcing other peptides, including VIP, option migraines.

Fear is characterized by distinct behavioral and physiological responses that are essential for the survival of human species. conditioning (FC) serves as a valuable model studying acquisition, extinction, expression fear. The serotonin (5-hydroxytryptamine, 5-HT) system known to play significant role in emotional motivational aspects behavior, including fear learning expression. Accumulating evidence from both animal studies suggests brain regions involved FC, such amygdala, hippocampus, prefrontal cortex, possess high density 5-HT receptors, implicating crucial involvement aversive learning. Additionally, exploring gene polymorphisms have indicated their potential influence on FC. Therefore, objective this work was review existing linking with memory humans. Through comprehensive screening PubMed Web Science databases, 29 relevant were included final review. These investigated relationship between using drug manipulations or 5-HT-related polymorphisms. results suggest elevated levels enhance learning, indicating modulation 5-HT2A receptors regulates Understanding neurochemical messenger associative can provide insights into psychiatric disorders anxiety post-traumatic stress disorder (PTSD), among others.
