Ageing is a complex and unavoidable physiological process which, in simple terms, consists of progressive deterioration the functionality cells, tissues organs, culminating an increased risk developing chronic pathologies. Telomeres, repetitive nucleotide structures at end chromosomes, ensure genomic integrity modulate cellular senescence. The shortening telomere length with each cell division directly correlates susceptibility to However, this shortening, normally inevitable, can be markedly accelerated presence infections, such as HIV-1 infection, by sustained continuous activation immune system, inflammation, generation oxidative stress, or direct alterations produced viral proteins. Thus, narrative review, we discuss 12 hallmarks ageing context understanding molecular changes induced through these well-established pillars could provide holistic approach management HIV-positive patients. At same time, considering that telomeres are centre all changes, assessment impact antiretroviral therapy on necessary guide clinical decisions. ultimate goal research develop personalised therapies increase quality life health outcomes HIV

Abstract Major depressive disorder (MDD) and substance-use disorders (SUDs) often lead to premature aging, increasing vulnerability cognitive decline other forms of dementia. This study utilized advanced systems bioinformatics identify aging “signatures” in MDD SUDs evaluated the potential for known lifespan-extending drugs target reverse these signatures. The results suggest that inhibiting transcriptional activation FOS gene family members holds promise mitigating SUDs. Conversely, antidepressant activating PI3K/Akt/mTOR pathway, a common mechanism rapid-acting antidepressants, may accelerate patients, making them unsuitable those with comorbid aging-related conditions like dementia Alzheimer’s disease. Additionally, this innovative approach identifies anti-aging interventions such as Deferoxamine, Resveratrol, Estradiol valerate, natural compounds zinc acetate, genistein, ascorbic acid, regardless anxiety disorders. These findings illuminate effects offer insights into treatment strategies patients conditions, including

Premature vascular aging and endothelial cell senescence are major risk factors for cardiovascular diseases atherothrombotic disturbances, which main complications of both acute long COVID-19. The S protein SARS-CoV2, acts as the receptor binding viral infection, is able to induce cells inflammation it has been found an isolated element in circulation human tissues reservoirs months after infection. Here, we investigated whether directly deciphered some mechanisms involved. In primary cultures umbilical vein (HUVEC), SARS-CoV-2 enhanced a concentration-dependent manner cellular content DNA damage response markers (senescence-associated-β galactosidase, γH2AX), well growth-arrest effectors (p53, p21, p16). parallel, reduced availability cytoprotective proteins, such anti-aging klotho, Nrf2 or heme oxygenase-1, caused functional harm by impairing ex vivo endothelial-dependent vasorelaxation murine microvessels. These effects were prevented pharmacological inhibition NLRP3 inflammasome with MCC950. Furthermore, supplementation either recombinant klotho angiotensin-(1-7), equally protected against pro-senescence, pro-inflammatory pro-oxidant action protein. Globally, this study proposes novel disease context COVID-19 its sequelae provides clues order prevent complications.

Aging is a complex biological process characterized by gradual deterioration in physiological activities, contributing to an elevated risk of different age-associated malignancies, including cancer. The current review paper aims elucidate the association between hallmarks aging and parallel development Various cellular molecular mechanisms underlying such as senescence, genomic instability, telomere shortening play significant role accumulation genetic mutations disruption activities. It has also discussed immune system’s aging, indicating how age-linked dysfunction compromises body’s capacity recognize eradicate pre-tumorous cells, thus promoting cancer advancement. This examines affects tumor microenvironment, where age-related alterations stromal extracellular matrix factors lead conditions that promote growth proliferation. Furthermore, it discusses concept “accelerated aging” survivors, highlighting negative effects its therapy intensify process, resulting greater physical cognitive decline. explores prospect new therapies target aging-related processes ameliorate outcomes. Overall, underscores need for ongoing interdisciplinary research understand relationship cancer, ultimately aiming develop more effective prevention treatment approaches.

Fanconi anemia, a rare disorder with an incidence of 1 in 300,000, is caused by mutations FANC genes, which affect the repair DNA interstrand crosslinks. The disease characterized congenital malformations, bone marrow failure within first decade life, and recurrent squamous cell carcinomas oral cavity, esophagus, anogenital regions starting around age 20. In this review, we propose that anemia should be considered premature-aging syndrome. Interestingly, onset severity life-limiting clinical features can influenced lifestyle choices, such as healthy diet physical activity. These factors shape epigenetic status at-risk types enhance competence immune system through nutritional signaling. may serve model for understanding aging process general population, addressing research gaps its presentation suggesting prevention strategies. Additionally, will discuss how balance genetic environmental risk factors—affecting both cancer speed aging—is interlinked signal transduction dietary molecules. underlying nutrigenomic principles offer guidance individuals well population.

Abstract Background The process of aging is an intrinsic and inevitable aspect life, affecting every organism. Diseases associated with pose a significant challenge to global healthcare system. In recent years, the application peptide therapy in geriatric research, anti-aging peptides have emerged as promising strategy for combating aging. These peptides, their low immunogenicity ease synthesis, hold promise potential therapeutics Consequently, accurate identification discovery more paramount importance. However, date, no computational methods been employed by researchers identify such peptides. Results this study, we innovatively constructed benchmark dataset based on biological functions significance these Subsequently, proposed Antiaging-FL, prediction model feature representation learning machine learning, predict results indicate that Antiaging-FL achieved AUC, ACC, MCC value 1.0, 0.997, 0.995, respectively, AAP400 dataset, 0.99, 0.97, 0.95, test set, demonstrating excellent predictive performance. Conclusions We developed powerful bioinformatics tool, expedite new putative future, could assist related fields provide insights into functional mechanisms while also guiding drug development research.
