Gastrodin is an extract from the dried tuber of Chinese herb Gastrodia elata (Tian ma), with anti-inflammatory, antioxidant, and antiviral properties. Recent studies have shown that, compared to commonly used diabetes drugs, gastrodin has antidiabetic effects in multiple ways, characteristics low cost, high safety, less side effects, protection β-cell function, relieving insulin resistance alleviating complications. In addition, it confirmed that can protect function lung other organs, enhance activity via upregulating type I interferon (IFN–I), inhibit angiotensin II (AngII), a key factor "cytokine storm" caused by COVID-19. Therefore, we reviewed effect mechanism on 2 mellitus (T2DM), speculated potential mechanisms signal pathway, inflammation, mitochondrial endoplasmic reticulum its prevention treatment We hope provide new direction strategy for basic research clinical work: considered as drug

Background: Cardiac risk rises during acute SARS-CoV-2 infection and in long COVID syndrome humans, but the mechanisms behind COVID-19-linked arrhythmias are unknown. This study explores term effects of on cardiac conduction system (CCS) a hamster model COVID-19. Methods: Radiotelemetry conscious animals was used to non-invasively record electrocardiograms subpleural pressures after intranasal infection. cytokines, interferon-stimulated gene expression, macrophage infiltration CCS, were assessed at 4 days weeks post-infection. A double-stranded RNA mimetic, polyinosinic:polycytidylic acid (PIC), vivo vitro activate viral pattern recognition receptors absence Results: COVID-19 induced pronounced tachypnea severe dysfunction, spanning from bradycardia persistent atrioventricular block, although no protein expression detected heart. Arrhythmias developed rapidly, partially reversed, then redeveloped pulmonary resolved, indicating CCS injury. Increased remodeling accompanied electrophysiological abnormalities. Interestingly, arrhythmia phenotype reproduced by injection PIC virus, that innate immune activation sufficient drive response. also strongly cytokine secretion robust interferon signaling hearts, human iPSC-derived cardiomyocytes (hiPSC-CMs), engineered heart tissues, alterations electrical Ca2+ handling properties. Importantly, blunted inhibition JAK/STAT or mitochondrially-targeted antioxidant. Conclusions: The findings indicate dysfunction cell is COVID-19, arising indirectly oxidative stress excessive responses infection, with implications for Syndrome.

The Spike protein of SARS-CoV-2 acts as an enterotoxin able to induce chloride secretion and production reactive oxygen species (ROS), involved in diarrhea pathogenesis. L. rhamnosus GG (LGG) is recommended pediatric acute gastroenteritis guidelines a therapy independent infectious etiology. We tested postbiotic preparation LGG (mLGG) vitro model COVID-associated diarrhea. Caco-2 cell monolayers mounted Ussing chambers were exposed protein, electrical parameters secretory effect (Isc TEER) recorded the system. Oxidative stress was analyzed by measuring ROS (DCFH-DA), GSH levels (DNTB), lipid peroxidation (TBARS). Experiments repeated after mLGG pretreatment cells. Isc increase induced consistent with pattern, which dependent on oxidative defined 2-fold variation glutathione levels. significantly reduced (p = 0.002) stress, namely < 0.001), level changes 0.001). counteracts Spike-induced inhibiting enterotoxic stress. efficacy form depends metabolites secreted medium antioxidant properties similar NAC.
