The aim. To determine the role and place of endothelial dysfunction in pathogenesis comorbid gastroesophageal reflux disease (GERD) obesity by studying levels nitric oxide (NO) metabolites blood urine. evaluate efficacy benefits prescribed therapy, which included additional inclusion a peptide compound analogous to leu-enkephalin tyrosine-2-alanine-glycine-phenylalanine-leucyl-arginine diacetate (Dalargin). Materials methods. Clinical manifestations were assessed using GERD-HRQL questionnaire, stable NO2 total + NO3 determined spectrophotometric method, leptin enzyme-linked immunosorbent assay, pH-metry motor function was studied ultrasound. 130 patients examined, including 70 with GERD first degree, 40 without concomitant pathology 20 healthy individuals. study divided into 3 groups. Results. It that NO2, urine group are significantly lower than isolated (p<0.001). A direct correlation between level acidity stomach oesophagus established; an inverse relationship NO degree gastrointestinal functional disorders, severity clinical manifestations, hormonal secretion adipocytes. supplemental Dalargin improved effectiveness quality pathogenetic therapy achieved more positive dynamics is recommended for prescription practice. Conclusions. Obesity predictor severe course dysfunction. There functional, secretory disorders oesophagus.

Nitric oxide (NO) has been defined as the “miracle molecule” due to its essential pleiotropic role in living systems. Besides implications physiologic functions, it is also involved development of several disease states, and understanding this ambivalence crucial for medicinal chemists develop therapeutic strategies that regulate NO production without compromising beneficial functions cell physiology. Although nitric synthase (NOS), i.e., enzyme deputed biosynthesis, a well-recognized druggable target bioavailability, some issues have emerged during past decades, limiting progress NOS modulators clinical trials. In present review, we discuss most promising advancements research small molecules are able activity with improved pharmacodynamic pharmacokinetic profiles, providing an updated framework field could be useful design new modulators.

Polyphenols are dietary bioactive compounds, many of which have anti-inflammatory properties. However, information on the intake polyphenols at class and compound levels their associations with gastrointestinal (GI) systemic inflammation is lacking. Estimate polyphenol in healthy adults examine its relationship GI markers. Healthy (n = 350) completed United States Department Agriculture Nutritional Phenotyping Study, an observational, cross-sectional study balanced for age, sex, body mass index. Dietary intake, assessed via multiple 24-h recalls, was ingredientized mapped to FooDB, a comprehensive food composition database. (total, class, compound) estimated examined markers using linear models random forest regressions. Mean total ∼914 mg/1000 kcal/d flavonoids as greatest contributor (495 kcal/d). Tea, coffee, fruits were among largest contributors intake. Total negatively associated marker, fecal calprotectin (β −0.004, P 0.04). At level, categorized prenol lipids −0.94, < 0.01) phenylpropanoic acids −0.92, plasma lipopolysaccharide-binding protein, proxy permeability. Food sources these 2 classes included mainly olive products. We further detected positive association between C-reactive protein "cinnamic derivatives" hierarchical feature engineering modeling. Even adults, Relationships inflammatory outcomes varied resolution—total, compound—of suggesting nuanced impact inflammation. This trial registered clinicaltrials.gov NCT02367287.

Individuals born after intrauterine growth restriction (IUGR) are at risk of developing cardiovascular diseases (CVDs). Endothelial dysfunction plays a role in the pathogenesis CVDs; and endothelial colony-forming cells (ECFCs) have been identified as key factors repair. In rat model IUGR induced by maternal low-protein diet, we observed an altered functionality ECFCs 6-month-old males, which was associated with arterial hypertension related to oxidative stress stress-induced premature senescence (SIPS). Resveratrol (R), polyphenol compound, found improve function. this study, investigated whether resveratrol could reverse ECFC dysfunctions group. were isolated from control (CTRL) males treated R (1 μM) or dimethylsulfoxide (DMSO) for 48 h. IUGR-ECFCs, increased proliferation (5′-bromo-2′-deoxyuridine (BrdU) incorporation, p < 0.001) improved capillary-like outgrowth sprout formation (in Matrigel), nitric oxide (NO) production (fluorescent dye, 0.01), synthase (eNOS) expression (immunofluorescence, 0.001). addition, decreased reduced superoxide anion 0.001); Cu/Zn dismutase (Western blot, 0.05); reversed SIPS beta-galactosidase activity (p 0.001), p16ink4a 0.05) Sirtuin-1 expressions blot). No effects CTRL-ECFCs. These results suggest that reverses long-term IUGR.
