Circadian rhythm disruption (CRD), stemming from sleep disorders and/or shift work, is a risk factor for reproductive dysfunction. CRD has been reported to disturb nocturnal melatonin signaling, which plays crucial role in female reproduction as circadian regulator and an antioxidant. The hypothalamic-pituitary-ovarian (HPO) axis regulates reproduction, with luteinizing hormone (LH) pulse pattern playing pivotal folliculogenesis steroidogenesis. However, the effect of on HPO involvement remains unclear. Female CBA/CaJ mice underwent modeling, involves alternating between standard light conditions 8-h advance schedule every 3 days 8 weeks, whereas control were maintained under 12:12-h light/dark (LD) cycle. Premature ovarian insufficiency (POI) is a diverse form of female infertility characterized by decline in function before the age 40. Melatonin (MT) potential clinical treatment for restoring or safeguarding POI. However, specific therapeutic mechanism underlying this effect remains unclear. To address this, we conducted experiments using human granulosa cells (GCs) from both POI and normal patients. We examined expression levels autophagy-related genes proteins GCs through qRT-PCR western blot analysis. Autophagy flux was monitored infected with GFP-LC3-adenovirus, regulatory MT autophagy investigated. Additionally, employed pharmacological intervention 3-Methyladenine (3-MA) RNA interference Forkhead box O-3A (FOXO3A) to elucidate process. Oocyte aging is closely related to a decline in female fertility, accompanied by increased reactive oxygen species levels and changes protein posttranslational modifications. However, the role of palmitoylation oocyte has not been investigated. In present study, new association between redox oocytes was found. We found that level palmitoyl-protein thioesterase 1 (PPT1), depalmitoylation enzyme, maternally aged mice follicular fluid (age >35 years) patients with decreased ovarian reserve (DOR). Elevated PPT1 led S-palmitoylation oocytes, which impaired maturation spindle formation. Tubulin identified as critical palmitoylated regulated PPT1, whose also advanced age, abnormalities membrane localization microtubule polymerization. Melatonin down-regulate excessive rescue PPT1-induced damage mouse only regulating oxidative stress, but binding regulate its lysosomal degradation. summary, our data demonstrate participates tubulin palmitoylation, providing evidence stress regulates revealing novel mechanism aging.
