Oxidative stress, associated with excessive production of reactive oxygen and nitrogen species (ROS, RNS), contributes to the development progression many ailments, such as aging, cardiovascular diseases, Alzheimer's disease, Parkinson's diabetes, cancer, preeclampsia or multiple sclerosis. While phenols polyphenols are most studied antioxidants structurally similar compounds anilines thiophenols sporadically analyzed despite their radical scavenging potential. This work assesses impact structural features on antioxidant activity. Seventeen pairs phenol/thiophenol analogues, possessing both electron-donating withdrawing groups were selected for this study. Several physicochemical properties determined by density functional theory (DFT) calculations at (U)B3LYP/6-311++G(d,p) level gas phase scrf = (smd, solvent water) solvated ones. Correlations between calculated experimental activities investigated identify pivotal physical characteristics contributing efficiency. These include S–H O–H bond distances dissociation enthalpies (BDE), dipole moments, logP values, highest occupied molecular orbital (HOMO) lowest unoccupied (LUMO) energies, HOMO–LUMO gap energies that M06-2X/6-311++G(d,p) theory, Fukui functions.
