Abies alba Mill. is a prominent European tree species predominantly inhabiting cool and humid montane environments. However, paleoecological evidence reveals that during the Eemian mid-Holocene, A. thrived in much warmer drier climates. This capacity nowadays reflected cryptic meso- sub-Mediterranean lowland populations. To link populations across diverse climates spanning from Mediterranean lowlands to Alpine timberline, we genotyped 421 specimens Italy Switzerland at 190 single-nucleotide polymorphisms (SNPs). Population genetic structure analyses indicate isolated Tuscany Ticino align genetically with higher elevation each region, suggesting their thrive warmer, conditions cannot be attributed plantations planting stock originating different or occurrence of single warm-adapted lineage showing disjunct distribution, unless migration between stands occurred. Genotype-environment associations reveal temperature seasonality, precipitation critical seasons, relative humidity are important for explaining species' variation. With genotype-environment genotype-phenotype associations, identified candidate adaptive genes potentially linked climatic drought response. While certain alleles may have spread Southern could explained by shared ancestry Tuscan populations, local adaptation occurred specific loci. These findings underscore importance considering hitherto overlooked better understand niche its potential forest conservation management under global warming.

Sugarcane (Saccharum spp.) hybrid, one of the most important crops in Florida, has been affected by orange rust (OR) disease caused Puccinia kuehnii since 2007, resulting significant yield loss. Developing resistant cultivars to this become an goal sugarcane breeding programs. However, specific genes and molecular mechanisms underlying resistance OR are still not clear. In study, we selected two sister lines with different genotypes—showing contrasting responses disease—from a major quantitative trait loci (QTL) region controlling resistance. Morphological anatomical observations revealed that line (540) had significantly smaller stomatal size lower density than susceptible (664). Transcriptomic analyses showed 540 increased cell surface modification activity, suggesting possible receptors. Differentially expressed gene coexpression also key involved biosynthesis anti-fungal molecules, such as hordatines, arabidopyrones, alkaloids. They strong increase long non-coding RNA expression, playing role transcriptional regulation. Transcriptomic–metabolomic joint analysis suggested phenylpropanoid derivatives purported antioxidant capabilities 540, especially those deriving from ferulate. Genes, pathways, some single-nucleotide polymorphisms identified study will provide fundamental information resources advance knowledge genetic relation disease, supporting programs developing improved OR.

Southern leaf blight (SLB), caused by Bipolaris maydis, poses a significant threat to maize and popcorn production. To understand the molecular mechanisms underlying SLB resistance, we conducted high-throughput proteomic analysis comparing SLB-resistant (L66) SLB-susceptible (L51) genotypes at four ten days after inoculation (DAI). A total of 717 proteins were identified, with 151 differentially accumulated (DAPs) between genotypes. Eighteen DAPs exhibited same regulatory pattern in both (R4/S4) (R10/S10) DAI. The protein-protein interaction (PPI) network linked resistance susceptibility enriched specific metabolic pathways response, including photosynthesis, ribosome, ascorbate aldarate metabolism, glutathione carbon metabolism. Proteins such as photosystem II 11 kD protein (B4FRJ4, PSB27-1), which was up-regulated time points (R4/S4 R10/S10), 60S acidic ribosomal P0 (A0A1D6LEZ7, RPP0B), unique resistant genotype (R4 R10), highlighted importance maintaining photosynthetic efficiency synthesis during pathogen attack. Additionally, dehydroascorbate reductase like-3 (B4F817, DHAR3) consistently genotypes, emphasizing its role redox balance ROS detoxification. In contrast, glyceraldehyde-3-phosphate dehydrogenase (K7UGF5, GAPC2), glycolytic enzyme, susceptible genotype, suggesting involvement managing energy metabolism under stress conditions. Our findings suggest that involves combination enhanced repair, homeostasis, activity, providing new potential targets, DHAR3 RPP0B, for genetic improvement resistance. These results offer valuable insights into breeding programs aimed developing varieties.

Flooding stress, including waterlogging and submergence, is one of the major abiotic stresses that seriously affects growth development plants. In present study, physiological, epigenetic, transcriptomic analyses were performed in wheat seedling leaves under (WL), half submergence (HS), full (FS) treatments. The results demonstrate FS increased leaves’ hydrogen peroxide (H2O2) malondialdehyde (MDA) contents reduced their chlorophyll (SPAD), photosynthetic efficiency (Fv/Fm), shoot dry weight more than HS WL. addition, catalase (CAT) peroxidase (POD) activities However, there no significant differences H2O2, MDA, SPAD, Fv/Fm, superoxide dismutase (SOD) POD between WL changes DNA methylation related to stress types, increasing treatments decreasing treatment. Additionally, a total 9996, 10,619, 24,949 genes differentially expressed WL, HS, treatments, respectively, among which ‘photosynthesis’, ‘phenylpropanoid biosynthesis’, ‘plant hormone signal transduction’ pathways extensively enriched three flooding involved these showed flooding-type-specific expression. Moreover, responses observed conditions, enrichment specific TFs response pathways. These will contribute better understanding molecular mechanisms underlying provide valuable genetic epigenetic information for breeding flood-tolerant varieties wheat.
