Electroacupuncture (EA) is a form of Traditional Chinese Medicine (TCM) that combines acupuncture with microcurrents mimicking the body's bioelectricity to prevent and treat diseases. Previous studies have demonstrated its antidepressant-like effects in chronic unpredictable mild stress (CUMS)-induced ovariectomy (OVX) rats. Neural stem cell-derived exosomes (NSCs-Exo) are heterogeneous targeted, effectively promoting nerve regeneration repairing neuronal damage, while potentially conveying EA. However, precise mechanism remains unclear. In this study, perimenopausal depressive disorder (PDD) rat model were established using two-step protocol CUMS + OVX. Treatment EA combined NSCs-Exo (EA-Exo) significantly improved depression-like behaviors PDD rats, as indicated by increased sucrose intake Sucrose Preference Test (SPT), reduced immobility Forced Swimming (FST), prolonged activity Out-of-Field (OFT). EA-Exo treatment increasing serum levels 5-hydroxytryptamine (5-HT) decreasing FST. It also alleviated OVX-CUMS-induced disturbances energy metabolism, inflammation, oxidative responses enhancing 5-HT, dopamine (DA), ATP, superoxide dismutase (SOD), interleukin-10 (IL-10), reducing cyclic AMP (cAMP), interleukin-6 (IL-6), reactive oxygen species (ROS), malondialdehyde (MDA). Furthermore, reversed structural functional impairments hippocampal synapses mitochondria. This was evidenced reductions synaptic plasticity proteins PSD95, SYN, GAP43, well decreased expression metabolism pathway AMPK, NRF1, PGC1α, TFAM. These findings suggest ameliorates behavior OVX-CUMS rats modulating activating AMPK/NRF1/PGC1α/TFAM signaling pathway.

Progestogens’ anti-anxiety and anti-depressive effects mechanisms are not well-understood. Progestogens hypothesized to have on behavior, independent of actions at nuclear progestin receptors (NPRs) dependent allopregnanolone (5α-pregnan-3α-ol-20-one; 3α,5α-THP), a 5α-reduced, neuroactive metabolite progesterone (P4). Adult c57 mice in behavioral estrus (proestrus; pro) showed more anti-anxiety-like anti-depressant-like behavior higher levels estradiol (E2), P4, the hippocampus/amygdala complex. Proestrus > 5α-reductase knockout (5αRKO) made central entries an open field than diestrus 5αRKO that were different. Ovariectomized (OVX) administered 1, 2, or 4 mg/kg P4 SC dosage-dependent increases (more behavior); had maximal 1–2 P4. OVX maximum with 3α,5α-THP (4 mg/kg), c57s similar response but 5αRKOs was half dosage. receptor (PRKO) decreased immobility (depression-like behavior) forced swim task. Effects E2 veh both groups. Levels consistent field. brain-derived neurotrophic factor (BDNF) greater among E2-primed (0.09 mg/kg, SC) vs vehicle-administered mice. In sum, adult female can be responsive for anti-anxiety/anti-depressant-like behavior; such may NPRs require 5α-reduction E2’s priming BDNF

When postmenopausal women are under stress conditions, this exacerbates mood disorders and issues with neuroimmune systems. The porcine placenta is known to relieve menopausal depression in clinical trials, but its underlying mechanisms for anti-inflammatory functions remain poorly defined. present study was designed examine the effects of enzymatic hydrolysate (EPPH) on LPS-induced levels nitric oxide (NO), prostaglandin E2 (PGE2), corticosterone (CORT), pro-inflammatory cytokine interleukin-1 beta (IL-1β) RAW 264.7 macrophage cells. In addition, neurite outgrowth PC12 cells evaluated EPPH growth. To mimic symptoms menopause-related depression, a stressed ovariectomized (OVX) female mouse model used evaluate antidepressant EPPH. mice were randomly divided into five groups: (1) sham-operated (Sham) group, (2) OVX + repeated saline-treated (OVX ST) (3) estradiol (0.2 mg/kg)-treated (positive control) (4) (300 (300) (5) (1500 (1500) group. Female repeatedly immobilization-stressed 2 weeks (2 h/day). A tail suspension test conducted 13th day, followed by forced swimming 14th day assess After behavioral tests, CORT, PGE2, IL-1β evaluated. c-Fos expression paraventricular nucleus (PVN) using immunohistochemistry. concentrations NO, stimulated LPS significantly reduced via addition promoted compared that controls. test, duration immobility treated 1500 ST group had decreased c-Fos-positive neurons PVN CORT serum Sham These results suggested high dose administration induced antidepressant-like effect downregulating IL-1β, PGE2 through reducing regions.
