Heavy metals are often found in soil and can contaminate drinking water, posing a serious threat to human health. Molecular pathways curation therapies for mitigating heavy metal toxicity have been studied long time. Recent studies on oxidative stress aging shown that the molecular foundation of cellular damage caused by metals, namely, apoptosis, endoplasmic reticulum stress, mitochondrial share same as those involved senescence aging. In recent studies, many types exposures used both animal models. Chelation therapy is traditional treatment toxicity. However, recently, various antioxidants be effective treating metal-induced damage, shifting research focus investigating interplay between metals. this review, we introduce basis its relationship with aging, summarize clinical implications, discuss other agents protective effects against damage.

Heavy metals like arsenic, mercury, cadmium, and lead are harmful pollutants that can change how our genes regulated without altering the DNA sequence, specifically through a process called methylation (DNAm) at 5-methylcytosine, an epigenetic mark we will focus on in this review. These changes DNAm most sensitive during pregnancy, critical time for development when these modifications affect traits expressed. Historically, research environmental effects has focused adults, but now there is more emphasis studying impacts early childhood. The placenta acts as protective barrier between mother baby, by examining it, scientists identify key might long-term health. Human exposure to heavy metals occurs through multiple pathways in daily life. However, the effects of mixed metal on accelerated aging elderly U.S. population remains unclear. This study aims investigate association between concentrations blood and urine onset aging. A total 2530 individuals were included analysis 716 this study. GrimAge acceleration (AgeAccelGrim) was calculated as residuals from regressing DNA methylation chronological age. Weighted multivariable logistic regression models applied analyze relationship with AgeAccelGrim. We are facing a critical aging crisis, with geriatric populations (65+) growing to unprecedented proportions and ~4 million people (a 6.5-fold increase) expected become centenarians by 2050. This is compounded environmental pollution, which affects individuals of all ages contributes age-related diseases. As we have limited understanding how pollutants affect older distinctly from younger populations, these longer-lived present key knowledge gap. To address this gap, employ “Toxic Aging Coin” approach: heads consider age impacts chemical toxicity, tails chemicals act as gerontogens—or they accelerate biological aging. employed approach investigate hexavalent chromium (Cr[VI]) on female rats exposed 0.05 or 0.1 mg Cr(VI)/L in drinking water for 90 days; the maximum contaminant levels (i.e., highest permitted) World Health Organization U.S. Environmental Protection Agency, respectively. During exposure, performed battery behavior assays assess grip strength, locomotor coordination, anxiety, spatial memory, sociability, social novelty preference. observed differences Cr(VI) neurotoxicity, function, memory middle-aged females being particularly affected. further compared results males, noting many sex differences, especially rats. Aging causes various degenerative diseases in the older adult population. Senescence, a state of permanent cell-cycle arrest accompanied by production pro-inflammatory factors known as senescence-associated secretory phenotype (SASP), is considered significant contributor to aging process and its chronic diseases. Ample evidence showed that stressors could induce senescence, including DNA damage, telomere shortening activation oncogenes, mitochondrial dysfunction. Numerous credible findings indicate environmental agents can UV radiation, high-fat diet, heavy metal exposure, cigarette smoke. These posed possibility many more may senescence accelerate but remain unidentified. Senescence also becomes intriguing due promising future pharmacological target blunt detrimental effects prevent aging-related diseases, either eliminating senescent cells or controlling SASP. On other hand, investigating has become intricate need for multi-marker approach translation vivo analysis. This review will discuss biomarkers, how identify gerontogens vivo, development senotherapy targets cells.
