Epilepsy is one of the most common neurological disorders globally, affecting about 50 million people, with nearly 80% those affected residing in low- and middle-income countries. It characterized by recurrent seizures that result from abnormal electrical brain activity, varying widely manifestation. The exploration biomechanical effects have on dynamics stress levels relevant for development more effective treatments protective strategies. This study uses a blend experimental data computational simulations to assess brain’s physical response during seizures, particularly focusing behavior cerebrospinal fluid resulting mechanical stresses different regions. Notable findings show increases stress, predominantly posterior gyri brainstem, an evidence displacement relative skull. These observations suggest dynamic complex interaction between skull, maximum shear regions demonstrating limited yet essential role CSF. Cannabinoids have shown potential in drug-resistant epilepsy treatment; however, we lack knowledge on which cannabinoid(s) to use, dosing, and their pharmacological targets. This study investigated (i) the anticonvulsant effect of Cannabidiol (CBD) alone (ii) combination with Delta-9 Tetrahydrocannabinol (Δ9-THC), as well (iii) serotonin (5-HT)1A receptor’s role CBD’s mechanism action. Seizure activity, induced by 4-aminopyridine, was measured extracellular field recordings cortex layer 2/3 mouse brain slices. The 10, 30, 100 µM CBD combined Δ9-THC evaluated. To examine action, slices were pre-treated a 5-HT1A receptor antagonist before An amount ≥30 exerted significant effects while 10 did not. However, low-dose (20:3 ratio) displayed significantly greater than either phytocannabinoid alone. Furthermore, blocking receptors application abolished effects. Thus, our results demonstrate efficacy that exerts its effects, at least part, through receptors. These could address drug-resistance providing insight into laying groundwork for further testing cannabinoids anticonvulsants.
