Cervical cancer is preceded by low-grade squamous intraepithelial lesions (LSIL) and high-grade (HSIL). Human papillomavirus (HPV) test a sensitive method for cervical screening, but it less specific compared with cytological examination, leading to overtreatment reduced patient compliance. Therefore, new detection methods that can improve the accuracy of screening are needed. In present study, exfoliated cell samples were collected from 228 Chinese individuals, including 114 healthy control 46 patients LSIL, 21 HSIL 47 cancer. The DNA methylation levels 12 cancer-related genes detected using quantitative multiplex methylation-specific PCR. All individuals divided into high- or low-risk groups. Patients assigned high-risk group, whereas controls LSIL group. ability predict risks was evaluated ROC curves predictive model risk constructed linear regression analysis. significantly higher all in HSIL, those normal Family sequence similarity 19 member A4 (FAM19A4), phosphatase actin regulator 3 (PHACTR3), somatostatin (SST), Zic family 1 (ZIC1), paired box (PAX1) zinc finger protein 671 (ZNF671) used construct prediction, specificity 89.6% sensitivity 95.0%. study demonstrated Also, distinguish HSCL low risk.

The MAL family of integral membrane proteins consists MAL, MAL2, MALL, PLLP, CMTM8, MYADM, and MYADML2. best characterized members are elements the machinery that controls specialized pathways traffic cell signaling. This review aims to help answer following questions about MAL-family genes: (i) is their expression regulated in cancer and, if so, how? (ii) What role do they play cancer? (iii) Might have biomedical applications? Analysis large-scale gene datasets indicated altered levels transcripts specific types. A comprehensive literature search provides evidence dysregulation protein function repurposing cancer. For probably for other genes family, primarily a consequence methylation, although copy number alterations also contribute varying degrees. scrutiny two sources information, published studies, reveals potential prognostic applications as biomarkers—for instance, MAL2 breast cancer, MALL pancreatic MYADM lung cancer—and uses. availability validated antibodies some sanctions use biomarkers routine clinical practice.

Abstract In alignment with the World Health Organization's strategy to eliminate cervical cancer, substantial progress has been made in treatment of this malignancy. Cervical largely driven by human papillomavirus (HPV) infection, is considered preventable and manageable because its well‐established etiology. Advancements precision screening technologies, such as DNA methylation triage, HPV integration detection, liquid biopsies, artificial intelligence‐assisted diagnostics, have augmented traditional methods nucleic acid testing cytology. Therapeutic strategies aimed at eradicating reversing precancerous lesions refined pivotal measures for disease prevention. The controversy surrounding surgery early‐stage cancer revolves around identifying optimal candidates minimally invasive conservative procedures without compromising oncological outcomes. Recent clinical trials yielded promising results development systemic therapies advanced cancer. Immunotherapies, immune checkpoint inhibitors (ICIs), antibody‐drug conjugates (ADCs), targeted therapy demonstrated significant effectiveness, marking a advancement management. Various combination validated, ongoing aim enhance outcomes through novel drugs optimized regimens. prospect first malignancy be eliminated now within reach. review, we provide comprehensive overview latest scientific insights, particular focus on managements various stages disease, explore future research directions

Cervical cancer ranks as the second most fatal among women in developing countries, trailing only behind breast cancer. Human Papillomavirus (HPV) infection, various contributors, stands a primary cause of cervical Overexpression DNA methyltransferase 1 (DNMT1) leads to hypermethylation Cell Adhesion Molecule (CADM1) and Suppressor Cytokine Signaling (SOCS1), consequently silencing these tumor suppressor genes (TSGs) epigenetically. In this study, we explored reversal aberrant methylation squamous cells, SiHa using concentration 5 µg/ml Datura metel ethanol- chloroform (E: C) leaf extract. Treating cells with extract for 72 hours consecutively six days, fragmentation study cell apoptosis was performed. The methylation-specific PCR analyzed patterns TSGs their subsequent fragmentation. results suggest that able also effectively reversed promoter hypermethylation, leading reactivation CADM1 SOCS1. Abstract Background At present, there are certain deficiencies in cervical cancer screening methods. Consequently, we aimed to establish a prediction model for precancerous lesions utilizing DNA methylation haplotype load (MHL) markers. Methods Three machine learning models, namely, random forest (RF), support vector (SVM), and naive Bayes (NB), were developed based on the performance of 47 regions 52 HR-HPV + cytology specimens. Additionally, external validation three models was conducted using 101 cytological samples. Results From initial samples, derived with respective area under curve (AUC) values 0.864, 0.867 0.847. Subsequently, phase involving RF demonstrated superior compared E6 protein detection p16Ki67 predicting intraepithelial neoplasia grade 2 (CIN2) above. Compared human papillomavirus (HPV) combined triage model, had PPV 100% an NPV 95.15% HPV16/18-positive subgroup. Within other high-risk-HPV-positive subgroups, 41.67% 92.36%, respectively. Conclusions Our findings suggest that markers multiple genes offer clear advantages CIN2 above cancer. Furthermore, enhancing method has potential further improve model.

ABSTRACT Background To investigate the screening value of a small‐molecule probe to assess methylation PAX1 in cervical cancer. Materials and Methods The diagnostic threshold grayscale values for lesions was assessed by plotting Receiver Operating Characteristic (ROC) curve subjects. Grayscale were significantly different among four groups ( p < 0.05). Compared with LSIL cervicitis groups, considerably higher found CA high‐grade squamous intraepithelial lesion (HSIL) (both Results differential ROC curves showed that Area Under Curve low‐grade (LSIL) 0.8724 (95% CI = 0.7762–0.9685, 0.0001), 1.0000 HSIL 0.5484 0.3826–0.7142, 0.5755), 0.7724 0.6016–0.9432, 0.0138). Conclusion small molecular has certain application differentiating type better efficacy distinguishing inflammatory precancerous from carcinogenesis, but less determining lesions.
