We investigated the effects of low-level Er:YAG laser irradiation on proliferation and alternations in early gene expression gingival fibroblasts. Mice primary fibroblasts were irradiated with an (1.8, 3.9, 5.8 J/cm2). Irradiation at 3.9 J/cm2 promoted cell without significant changes lactate dehydrogenase or Hspa1a expression. Three hours after J/cm2, Fn1 level was significantly increased. RNA-seq identified 15 differentially expressed genes between non-irradiated cells, some which belonged to immediate (IEGs). Kyoto Encyclopedia Genes Genomes pathway analysis indicated MAPK enhancement, set enrichment showed TGF-β signaling set. Introduction: Periodontitis is an infectious disease of periodontal tissue caused by the bacteria Porphyromonas gingivalis which can trigger formation free radicals. be treated using metronidazole but long‐term use cause negative effects. Natural ingredients such as robusta coffee husk used alternative treatment for periodontitis because its anti‐inflammatory, antioxidant, and antibacterial benefits. Periodontitis causes inflammatory destruction of tooth-supporting tissues; however, the complex mechanism underlying its etiology remains unclear. Cuproptosis is a type cell death caused by an imbalance in intracellular copper homeostasis that leads to excess copper. However, changes expression and biological function cuproptosis-related genes (CRGs) periodontitis are not yet fully understood. This study investigated comprehensive effects differentially expressed CRGs (DE-CRGs) on via bioinformatic analysis. Nine DE-CRGs were discovered using normal gingival samples, single-cell RNA sequencing data analyzed identify them diverse clusters. We then detected correlation between immune infiltration, factors, mitochondrial dysfunction, diagnostic efficacy, predicted drugs. Moreover, DE-CRG whole tissue human fibroblast line (HGF-1) confirmed content HGF-1 cells investigated. Objectives: This study aimed to evaluate potential combining sodium fluoride (NaF) with compounds enhance antibiofilm effects Method: We found that a urea derivative significantly enhances efficacy by promoting retention ions within cells. synergistic BPU NaF were confirmed through various assays, including checkerboard time-kill growth curve analysis. These findings further supported additional methods, transmission electron microscopy (TEM), silico simulations, gene overexpression studies. Results: suggest targeting ion membrane exporters could efficacy. When combined fluoride, 1,3-Bis [3,5-bis(trifluoromethyl)phenyl]urea (BPU) showed increased inhibiting reducing established biofilms. Porphyromonas gingivalis, as the main etiology of periodontitis, causes inflammation in periodontal tissue, which triggers immune response, fibroblast decline, and collagen destruction, generating attachment loss. Fibroblasts perform a fundamental role repair process tissue. This study examined potential cassava leaf extract increasing quantity fibroblasts density gingiva rats with periodontitis. A posttest-only control group was used this study. The experiment involved 24 male Wistar divided into four different groups: group, induced by P. gingivalis given aquadest, metronidazole, extract. Gingival tissue taken after euthanasia, histological preparations were made, observed. One-way analysis variance revealed that showed notable difference between each (p < 0.05), interestingly, there no significant metronidazole least test results > 0.05). Cassava has to increase periodontitis rat models.
