This review discusses the relationship between cellular senescence and radiation exposure. Given wide range of ionizing sources encountered by people in professional medical spheres, as well influence natural background radiation, question effect on biological processes, particularly aging remains highly relevant. The parallel radiation-induced reveals common aspects underlying these processes. Based recent scientific data, key points effects processes associated with aging, such genome instability, mitochondrial dysfunction, altered expression miRNAs, epigenetic profile, manifestation senescence-associated secretory phenotype (SASP), are discussed. Unraveling molecular mechanisms can make a valuable contribution to understanding genetic basis age-associated diseases context environmental

Oxidative stress plays a role in various pathophysiological diseases, including neurogenerative such as Alzheimer′s disease (AD), which is the most prevalent neuro-pathology aging population. has been reported to be one of earliest pathological alterations AD. Additionally, it was demonstrated that older adults, there loss free radical scavenging ability. The Nrf2 transcription factor key regulator antioxidant defense systems, but, with aging, both amount and transcriptional activity decrease. With available treatments for AD being poorly effective, reinforcing systems via pathway may way prevent treat To highlight predominant signaling defending against oxidative and, therefore, neurotoxicity, we present an overview natural compounds exert their own neuroprotective roles through activation pathway. This review opportunity promote holistic approach treatment need further refine development new potential Nrf2-targeting drugs.

Drug addiction is a complex disease affected by numerous genetic and environmental factors. Brain regions in reward pathway, neuronal adaptations, epigenetic interactions causing transcriptional enhancement or repression of multiple genes induce different phenotypes for varying duration. Addictive drug use causes alterations similarly changes induced environment can promote addiction. Epigenetic mechanisms include DNA methylation post-translational modifications like methylation, acetylation, phosphorylation, ubiquitylation, sumoylation, dopaminylation crotonylation histones, ADP-ribosylation. Non-coding RNAs also changes. This review discusses these above areas stresses the need exploring epidrugs as treatment alternative adjunct, considering limited success current strategies. Epigenome editing complexes have lately been effective eukaryotic systems. Targeted cleavage techniques such CRISPR-Cas9 system, CRISPR-dCas9 complexes, transcription activator-like effector nucleases (TALENs) zinc-finger (ZFNs) exploited targeted recognition anchoring platforms, fused with writer eraser proteins delivered transfection transduction methods. Efficacy seen various neuropsychiatric conditions initial results involving model organisms are remarkable. Epidrugs present promising

L-Ascorbic acid, commonly known as vitamin C, has been used not only for disease prevention and in complementary alternative medicine, but also anti-aging purposes. However, the scientific evidence is yet sufficient. Here, we review physiological functions of C its relationship with various pathological conditions, including our previous findings, discuss prospects application healthy longevity. In summary, levels are associated lifespan several animal models. Furthermore, clinical studies have shown that blood lower middle-aged older adults than younger adults. Lower observed conditions such chronic kidney obstructive pulmonary elderly. These observations suggest implications age-related mechanisms owing to functions.

ABSTRACT Aims This study aimed to delineate the effect of hyperglycemia on Alu/LINE‐1 hypomethylation and in ERK1/2 genes expression type 2 diabetes with without cataract. Methods included 58 diabetic patients cataracts, 50 36 healthy controls. After DNA extraction bisulfite treatment, LINE‐1 Alu methylation levels were assessed using Real‐time MSP. gene was analyzed through real‐time PCR. Total antioxidant capacity (TAC), fasting plasma glucose (FPG) measured colorimetric methods. Statistical analysis performed SPSS23, setting significance level at P < 0.05. Results The TAC significantly lower for cataract groups than controls (259.31 ± 122.99, 312.43 145.46, 372.58 132.95 nanomole Trolox equivalent) a significant correlation between FPG both ( 0.05). sequences found be statistically hypomethylated compared In these groups, directly correlated 0.05) but not LINE‐1. higher patients, showing increases 2.41‐fold 1.43‐fold ERK1, 1.27‐fold 1.5 ERK2, respectively. ERK1 levels. A reverse observed expression. Conclusions Our findings indicate that hyperglycemia‐induced oxidative stress may alter patterns induce aberrant sequences. These changes play contributing role complications such as cataracts.

Aging is an inevitable physiological process, but delaying aging has always been enduring human pursuit. Silibinin (SIL), derived from the seeds of milk thistle plant, exhibits a broad spectrum pharmacological properties, including anti-tumor effects, liver protection, inhibition apoptosis, and alleviation inflammation. However, whether it anti-aging effects remains unclear. The SIL dietary supplement to Drosophila melanogaster prolonged lifespan, improved climbing ability, ameliorated age-associated intestinal barrier disruption, enhanced resistance oxidative stress, increased enzyme activities superoxide dismutase (SOD) catalase (CAT). Furthermore, RNA-seq results showed that addition significantly upregulated 74 genes downregulated 50 compared with control. KEGG (Kyoto Encyclopedia genomes) analysis demonstrated these differentially expressed were primarily involved in Toll signaling pathway endoplasmic reticulum proteins processing, six among which, IM2, IM3, Drsl3, CG7556, GCS1, TRAM, particularly regulation by supplementation. indicate enhancing antioxidant capacity regulating aging-related pathways. Therefore, shows potential application regimens.

Environmental toxins and epigenetic changes have been linked to neurodegenerative diseases, including Alzheimer's Disease (AD), Parkinson's (PD), amyotrophic lateral sclerosis (ALS). This paper aimed (i) identify environmental associated with AD, PD, ALS, (ii) locate potential industrial sources of in the United States (U.S.), (iii) assess driven by exposure reported patients. factors biomarkers neurodegeneration were compiled from 69 studies literature using Preferred Reporting Items for Systematic Reviews Meta Analyses (PRISMA) geographic information system approaches. Some 127 or putatively four toxic metals (As, Cd, Mn, Hg) common all three these diseases. (e.g., DNA methylation) patients include air pollutants, metals, organic chemicals pesticides, mycotoxins, cyanotoxins). Geographic analysis showed that study locations U.S., Europe, East Asia) selected researchers based on convenience access rather than risk disease prevalence. We conclude several markers shared among diseases could serve as attractive future targets guiding quality improvements aiding early detection.
