Rheumatoid arthritis (RA) is an autoimmune disorder characterized by inflammation and bone erosion. The exact mechanism of RA still unknown, but various immune cytokines, signaling pathways effector cells are involved. Disease-modifying antirheumatic drugs (DMARDs) commonly used in treatment classified into different categories. Nevertheless, based on a "trial-and-error" approach, substantial proportion patients show failed therapy for each DMARD. Over the past decades, great efforts have been made to overcome failure, including identification biomarkers, exploration reasons loss efficacy, development sequential or combinational DMARDs strategies approval new DMARDs. Here, we summarize these efforts, which would provide valuable insights accurate clinical medication. While gratifying, researchers realize that far from enough recommend specific individual patients. Precision medicine emerging medical model proposes highly individualized tailored approach disease management. In this review, also discuss potential precision overcoming with introduction cutting-edge technologies big data.

Emerging evidence indicates that resolution of inflammation is a critical and dynamic endogenous process for host tissues defending against external invasive pathogens or internal tissue injury. It has long been known autoimmune diseases chronic inflammatory disorders are characterized by dysregulated immune responses, leading to excessive uncontrol inflammation. The dysregulation epigenetic alterations including DNA methylation, posttranslational modifications histone proteins, noncoding RNA expression implicated in the system. response considered as trigger turn intercede actions. Thus, understanding molecular mechanism dictates outcome targeting regulators disease required resolution. In this article, we elucidate role nuclear factor-κB signaling pathway, JAK/STAT NLRP3 inflammasome diseases. And formulate relationship between inflammation, coronavirus 2019, human cancers. Additionally, review involved innate cells. All matters propose discuss rejuvenation potential interventions target regulatory mechanisms inflammation-associated improve therapeutic outcomes.

Rheumatoid arthritis (RA) is a chronic autoimmune disease that can lead to joint damage and even permanent disability, seriously affecting patients’ quality of life. At present, the complete cure for RA not achievable, only relieve symptoms reduce pain patients. Factors such as environment, genes, sex induce RA. Presently, non-steroidal anti-inflammatory drugs, DRMADs, glucocorticoids are commonly used in treating In recent years, some biological agents have also been applied clinical practice, but most side effects. Therefore, finding new mechanisms targets necessary. This review summarizes potential discovered from perspective epigenetics mechanisms.

Endogenous DNA derived from the nuclei or mitochondria is released into bloodstream following cell damage death. Extracellular DNA, called cell-free (cfDNA), associated with various pathological conditions. Recently, multiple aspects of cfDNA have been assessed, including levels, integrity, methylation, and mutations. Rheumatoid arthritis (RA) most common form autoimmune arthritis, treatment RA has highly varied outcomes. in patients elevated peripheral blood synovial fluid disease activity. Profiling may then be utilized clinical practice, such as prediction prognosis responses; monitoring state; a diagnostic marker. In this review, we discuss RA, particularly sources correlation pathogenesis. We also highlight potential analyzing profiles to guide individualized approaches for RA.

Abstract Rheumatoid arthritis is an immune‐mediated inflammatory disease in which fibroblasts contribute to both joint damage and inflammation. Fibroblasts are a major cell constituent of the lining cavity called synovial membrane. Under resting conditions, have important role maintaining homeostasis, producing extracellular matrix lubricants. In contrast, during inflammation, pathology by pathogenic levels mediators that drive recruitment retention cells within joint. Recent advances single‐cell profiling techniques transformed our ability examine fibroblast biology, leading identification specific subsets, defining previously underappreciated heterogeneity disease‐associated populations. These studies challenging held dogma homogeneous providing unique insights into their pathology. this review, we discuss recent understanding how contributes rheumatoid arthritis. Finally, address these could lead development novel therapies directly target selective populations future.

DNA methylation is the most important epigenetic modification involved in regulation of transcription, imprinting, establishment X-inactivation, and formation a chromatin structure. genome often associated with transcriptional repression closed heterochromatin. However, results genome-wide studies pattern activity genes have nudged us toward reconsidering this paradigm, since promoters many remain active despite their methylation.
