The gut–brain axis (GBA) is a complex bidirectional communication network connecting the gut and brain. It involves neural, immune, endocrine pathways between gastrointestinal (GI) tract central nervous system (CNS). Perturbations of GBA have been reported in many neurodegenerative disorders (NDDs), such as Alzheimer’s disease (AD), Parkinson’s (PD), amyotrophic lateral sclerosis (ALS), among others, suggesting possible role pathogenesis. microbiota pivotal component GBA, alterations its composition, known dysbiosis, associated with dysfunction neurodegeneration. might influence homeostasis CNS by modulating immune and, more directly, regulating production molecules metabolites that systems, making it potential therapeutic target. Preclinical trials manipulating microbial composition through dietary intervention, probiotic prebiotic supplementation, fecal transplantation (FMT) provided promising outcomes. Immune system modulation is an intriguing part of scientific research. It well established that the immune plays a crucial role in orchestrating cellular and molecular key mediators, thus establishing powerful defense barrier against infectious pathogens. Gut microbiota represent complex community approximately hundred trillion microorganisms live mammalian gastrointestinal (GI) tract, contributing to maintenance gut homeostasis via regulation innate adaptive responses. However, impairment crosstalk between intestinal immunity may reflect on detrimental health issues. In this context, many studies have indicated probiotics their bioactive compounds, such as bacteriocins short chain fatty acids (SCFAs), display distinct immunomodulatory properties through which they suppress inflammation enhance restoration microbial diversity pathological states. Prebiotics have become an important functional food because of their potential for modulating the gut microbiota and metabolic activities. However, different prebiotics can stimulate growth probiotics. The optimization was focused on in this study order to representative probiotics' (Lacticaseibacillus rhamnosus (previously Lactobacillus rhamnosus) Bifidobacterium animalis subsp. lactis) function. culture medium supplemented with three prebiotics, including inulin (INU), fructooligosaccharides (FOS), galactooligosaccharides (GOS). All clearly probiotic strains both monoculture co-culture. specific rates L. B. lactis were shown GOS (0.019 h-1) FOS (0.023 h-1), respectively. prebiotic index (PI) scores INU (1.03), (0.86), (0.84) co-culture at 48 h significantly higher than control (glucose). mixture achieve high quality optimized using Box-Behnken design. optimum ratios INU, FOS, 1.33, 2.00, 2.67% w/v, respectively, highest stimulated occurring PI score (1.03) total short chain fatty acid concentration (85.55 µmol/mL). suitable ratio mixed will function as a ingredient foods or colonic foods.

Abstract To prepare functional yacon juice, this study investigates the optimal fermentation duration, shelf‐life, chemical composition, and flavor volatiles of juice fermented by Lactiplantibacillus plantarum QS7T capable utilizing fructo‐oligosaccharides (FOS), along with its effects on gut microbiota. The period was determined as 30 h, resulting in essential parameters: 3.46 g L −1 for total acids a viable lactic acid bacteria (LAB) count 2.3 × 10 8 CFU mL . predicted shelf life when stored at 4°C is 28 days. Throughout storage, maintained significant FOS LAB (8.0 7 ). Sargassum fusiforme alginate (SF-Alg) possess many pharmacological activities, including hypoglycemic and hypolipidemic. However, the mechanisms of SF-Alg remain unclear due to its low bioavailability. In this study, we evaluated therapeutic effect on high-fat diet (HFD)/streptozotocin (STZ)-induced type 2 diabetes (T2D) mice. intervention was found significantly reduce fasting blood glucose (FBG), triglycerides (TG), total cholesterol (TC), while increasing high-density lipoprotein (HDL-c) improving tolerance. addition, administrating diabetic mice moderately attenuated pathological changes in adipose, hepatic, heart tissues as well skeletal muscle, diminished oxidative stress. To probe underlying mechanisms, further analyzed gut microbiota using 16S rRNA amplicon sequencing, metabolites by non-targeted metabolomics. Here, increased some benign bacteria (Lactobacillus, Bacteroides, Akkermansia Alloprevotella, Weissella Enterorhabdus), decreased harmful (Turicibacter Helicobacter). Meanwhile, dramatically branched-chain amino acids (BCAAs) aromatic (AAAs) colon T2D mice, suggesting a positive benefit an adjvant agent for T2D.
