Introduction Nanodiamond (ND) refers to diamond particles with sizes from few near 100 nanometers. For its superb physical, chemical and spectroscopic properties, it has been proposed studied the aims for bio imaging drug delivery. Many modalities on conjugating molecules ND form ND-X more efficient delivery have demonstrated in cellular animal models.Area covered novel approaches utilizing nanodiamond as a platform recently. This review summarizes recent developments facilitated delivery, complexes preparations tests models. The outlook clinical translation is discussed.Expert opinion (ND-X) produced different methods are realized delivery; almost all studies reported being compared pure alone. However, of particle size less than 10 nm found toxic due surface structure, strongly aggregate. In vivo demonstrate accumulation organs no confirmed long-term effect their release available. Standardized materials needed advance applications level.

Abstract Two‐photon excitation (2PE) represents substantial advantages in biophotonics over traditional one‐photon (1PE), offering enhanced spatial resolution and deeper tissue penetration capabilities. Since its introduction the 1990s, two‐photon excited microscopy has spurred rapid emergence of novel 2PE‐based applications, including photodynamic therapy (PDT) targeted release biologically active agents. This review first elucidates fundamental principles 2PE before comprehensively examining evolution photosensitizers (PSs) for PDT (2P‐PDT), covering both nanomaterials small molecules, categorized based on their respective characteristics. Additionally, it explores advancements PS development integrating 2P‐PDT with other therapeutic modalities (e.g., photothermal therapy, chemotherapy, immunotherapy, gene etc.). Subsequent sections provide an overview photoactivatable compounds absorption properties controlled various agents drugs, genes, gasotransmitters), emphasizing potential anticancer applications. Last, a deep discussion deciphers prospects challenges 2P‐mediated cancer treatment, particularly concerning clinical translation.

Triple-negative breast cancer (TNBC) is an aggressive subtype of characterized by the lack estrogen receptor (ER), progesterone (PR), and human epidermal growth factor 2 (HER2). Chemotherapy remains primary treatment option, yet TNBC frequently develops resistance, leading to relapse metastasis. Emerging evidence highlights potential combining DNA methylation inhibitors with immune checkpoint (ICIs). contributes escape silencing immune-regulatory genes, thereby reducing tumor's visibility cells. Reversing this epigenetic modification can reinvigorate surveillance enhance efficacy immunotherapies. This review discusses role in progression evasion, focusing on recent advances combination therapies involving ICIs. We discuss underlying mechanisms that enable these therapeutic synergies, preclinical clinical supporting approach, challenges posed tumor heterogeneity, drug toxicity. Finally, we explore for personalized strategies incorporating multi-omics data optimize outcomes. The integration immunotherapy offers a promising avenue improving survival patients.
