Curcumin (CUR) is a natural compound that can be combined with miconazole (MCZ) to improve vulvovaginal candidiasis (VVC) caused by Candida albicans treatment’s efficacy. This study aimed develop ureasil–polyether (U-PEO) vaginal ovules loaded CUR and MCZ for the treatment of VVC. Physicochemical characterization was performed thermogravimetry (TGA), differential thermal analysis (DTA), Fourier transform infrared spectroscopy (FTIR), in vitro release. Antifungal assays were used determine minimum inhibitory concentrations (MICs) synergism between MCZ, activity U-PEO microdilution agar diffusion. TGA results showed high stability hybrid ovules. In DTA, amorphous character possible interaction observed. FTIR no chemical incompatibility drugs. release resulted 80% 95% released within 144 h. The MICs 256 2.5 µg/mL, respectively. Antifungal agents are not always efficient in resolving vulvovaginal candidiasis (VVC), a common genital infection caused by the overgrowth of Candida spp., including albicans, or preventing recurrent infections. Although lactobacilli (which dominant microorganisms constituting healthy human vaginal microbiota) important barriers against VVC, Lactobacillus metabolite concentration needed to suppress VVC is unknown.We quantitatively evaluated concentrations determine their effect on 27 strains crispatus, L. jensenii, gasseri, Lacticaseibacillus rhamnosus, and Limosilactobacillus vaginalis, with inhibitory abilities biofilms C. albicans clinical isolates.Lactobacillus culture supernatants suppressed viable fungi approximately 24%-92% relative preformed biofilms; however, suppression differed among species. A moderate negative correlation was found between lactate production biofilm formation, but no observed hydrogen peroxide formation. The poor solubility of clotrimazole in the aqueous medium and uncontrolled removal drug-loaded suppository content limit its effectiveness treatment vulvovaginal candidiasis. Reptiles may act as reservoirs or spreaders of potential pathogenic microorganisms including Candida yeasts. While the epidemiology yeast species has been thoroughly studied, virulence profile isolated is not well investigated. Therefore, this study aimed to assess haemolytic, phospholipase, lipase activities and biofilm formation yeasts from cloacal swabs venomous snakes Marrakech, Morocco (Group I, n = 40) non-venomous Cocullo, Italy II, 32). All Group 1 showed low production (Lz ≥ 0.90) haemolysin (Hz 0.90), only 35% them were phospholipase (Pz) producers (Pz > 0.90). In contrast, all 2 produced enzymes more than 62% high amounts ≤ 0.64; Lz 0.69; Hz 0.69). Data show that able produce factors, which vary according hosts their origin, thus suggesting role in harboring spreading environment. Since was lower ones, we discussed it be affected by venom composition. This will pave way for fungal infection control, alternative antifungal drugs order overcome resistance phenomena.

Background Despite the pathogenesis of vulvovaginal candidiasis (VVC) is multifactorial, this study aimed to assess whether phenotypic characteristics, such as biofilm production and quality, along with clinical symptoms, are associated recurrent VVC (RVVC). Methods Over 1 year (Oct 2021–Oct 2022), we prospectively included 271 patients ≥18 years who attended our institution, had Candida spp. isolated in vaginal swabs, provided informed consent. Patients were followed for year. isolates tested by following techniques: crystal violet (CV) biomass quantification, XTT metabolic activity microscopy area quantification. Clinical microbiological data also collected. Results Overall, 55 (20.3%) experienced at least one recurrence, 19 (7.0%) meeting criteria RVVC (≥3 episodes/year), 65 episodes total. Demographic characteristics similar both groups. Most C. albicans (90.0%). Median (interquartile, [IQR]) absorbance values CV 18/19 238/252 non-RVVC follows: CV, 1.850 (1.578–2.156) vs. 1.426 (1.081–1.823), p = 0.005; XTT, 0.184 (0.116–0.293) 0.228 (0.147–0.331), 0.253. (IQR) occupation percentage 16/19 16/252 was, respectively: 13.15 (8.54–16.9) 10.73 (5.88–17.73), 0.710. Conclusion was high production. Additionally, exhibited a tendency toward lower activity, which may contribute treatment failure.
