Hormone receptor-positive (HR+), HER2-negative metastatic breast cancer (mBC) remains a prevalent and challenging disease. Endocrine therapy (ET) combined with CDK4/6 inhibitors is the first-line standard of care, yet resistance mechanisms, including ESR1 mutations, drive disease progression. Novel oral selective estrogen receptor degraders (SERDs) have emerged as promising therapeutic agents after progression secondary to mutations. However, available studies on SERDs differ in design, study population, outcomes, necessitating critical review data. This explores clinical efficacy, safety profiles HR-positive, mBC, particularly following inhibitors. Recent key trials, EMERALD, SERENA-2, EMBER-3 AMEERA-3, are analyzed, highlighting their efficacy overcoming resistance, especially ESR1-mutant populations. Oral offer enhanced bioavailability convenience compared fulvestrant, representing advancement endocrine therapy. Their integration into treatment strategies, combination regimens ctDNA-driven approaches, may improve patient outcomes address mechanisms. other than refinement for selection limited. Further trials needed optimize SERD use define most effective strategies SERDs.

This study aimed to assess global, regional, and national trends in breast cancer burden attributable high body mass index (BMI) from 1990 2021 provide projections up 2036 using data the Global Burden of Disease (GBD) study. Systematic analysis GBD dataset. We extracted BMI-related dataset, covering 204 countries territories. Age-standardized mortality rates (ASMR), disability-adjusted life years (DALYs), estimated annual percentage changes (EAPCs) were calculated analyze temporal patterns regional differences. Future projected Bayesian age-period-cohort (BAPC) model. In 2021, BMI contributed 44,707 deaths 1,041,309 DALYs globally, reflecting a 138.5 % 142.7 increase, respectively, compared 1990. While global age-standardized DALY remained relatively stable, absolute significantly increased, particularly low- middle-SDI regions. High-SDI regions exhibited highest rates, but low-SDI showed fastest growth. Projections indicate that due will continue rise until 2036, driven by increasing obesity insufficient healthcare resources. High is major driver burden, showing not only high-income more prominently Effective strategies, including public health interventions targeting management, are needed control rising burden.

Abstract Breast cancer is the most commonly diagnosed worldwide. Despite advancements in therapeutic modalities, its prognosis remains poor owing to complex clinical, pathological, and molecular characteristics. Sonodynamic therapy (SDT) a promising approach for tumor elimination, using sonosensitizers that preferentially accumulate tissues are activated by low-intensity ultrasound produce reactive oxygen species. However, clinical translation of SDT faces challenges, including limited efficiency resistance posed microenvironment. The emergence nanomedicine offers innovative strategies address these obstacles. This review discusses enhancing efficacy sonosensitizers, rational structural modifications, improved tumor-targeted enrichment, microenvironment remodeling, imaging-guided therapy. Additionally, SDT-based multimodal therapies, such as sono-chemotherapy, sono-immunotherapy, sono-photodynamic therapy, their potential applications breast treatment summarized. underlying mechanisms briefly outlined. Finally, this highlights current challenges prospects SDT, providing insights into future may improve outcomes cancer. Graphical abstract

The metabolism of drugs and foreign substances in humans typically involves multiple enzymatic steps, particularly phase-1 biotransformation the liver, where various cytochrome P450 monooxygenases (CYPs) play crucial roles. This complexity can lead to a wide range metabolites. Understanding contributions individual CYPs their interactions within these intricate enzyme cascades be challenging. We recently developed an vitro platform employing Chinese Hamster Ovarian (CHO) cell clones. These clones express human oxidoreductase (CPR), each is defined by specific CYP expression, thus exhibiting no detectable endogenous activity (mono-CYP CHO platform). In this study, we investigated whether mono-CYP suitable tool for modeling complex drug metabolization reactions vitro. Tamoxifen (TAM) was selected as model substance due its role prodrug widely used breast cancer therapy, main active metabolite, endoxifen, arises from two-step primarily involving system. Specifically, combined CYP3A4 CYP2D6 believed essential efficient endoxifen production.
