Introduction Hyperbaric oxygen (HBO2) therapy involves the inhalation of pure in a pressure chamber under increased ambient pressure. Recent research indicates that circulating small extracellular vesicles (sEVs) play important roles human physiology and pathology. Therefore, objective this pilot study was to monitor impact HBO2 on levels sEVs serum patients with necrotizing soft-tissue infections (NSTI), aseptic bone necrosis (ABN) or idiopathic sudden sensory neural hearing loss (ISSNHL). Material methods Serum-derived were isolated quantified 80 before after applied for NSTI, ISSNHL ABN as well normal controls who received neither nor steroids. Results We observed significant increase (p < 0.05), significantly elevated compared NSTI 0.05) 0.01). Conclusions The may be evidence both intended reduction inflammation result steroid inhibitory effect oxidative stress induced by therapy. Thus, released during might an biological role mediating response promising approach gain further insights into therapeutic efficacy

Background: Traumatic brain injury (TBI) occurs in individuals of all ages, predominantly during sports, accidents, and active military service members. Chronic consequences TBI include declined cognitive motor function, dementia, emotional distress. Small extracellular vesicles (sEVs), previously referred to as exosomes, are nano-sized lipid that play a role intercellular communication. Our prior research established the efficacy sEVs derived from human adipose stem cells (hASC sEVs) accelerating healing injuries. The hASC biologic therapeutic need be stored at −20 °C or −80 °C. This limits their use translating everyday clinics inclusion first-aid kits for application immediately after injury. To address this, here we demonstrate can room temperature (RT) two months post lyophilization. Methods: A transmission electron microscope (TEM) nanoparticle tracking analysis (NTA) were used validate morphology lyophilized RT sEVs. Using vitro models neuronal mimicking physical injury, inflammation, oxidative stress, viable promote Results: maintain purity, size, upon rehydration. Lyophilized, showed better compared with Conclusions: storage maintains accelerate injuries cells. will advance sEVs, bringing them closer clinics, home kits, on battlefields by

To investigate the epigenetic footprint of idiopathic inflammatory myopathies (IIM) through characterization circulating extracellular vesicles (EVs) and expression EV-derived small non-coding RNAs (sncRNAs). In this cross-sectional study, EVs were isolated by size-exclusion chromatography from plasma patients with IIM age- sex-matched healthy donors (HD). sncRNAs sequenced quantified using Next-Generation Sequencing (NGS). Following quality control normalization, filtered count reads used for differential microRNA (miRNA) piwi-interacting RNA (piRNA) analyses.
