Abstract Extracellular vesicles (EVs) are key in intercellular communication, carrying biomolecules like nucleic acids, lipids, and proteins. This study investigated postprandial characteristics proteomic profiles of circulating large extracellular (lEVs) healthy individuals. Twelve participants fasted overnight before baseline assessments. After consuming a controlled isocaloric meal, lEVs were isolated for flow cytometric analysis. Plasma triacylglyceride (TAG) levels confirmed fasting completion, while protein concentrations plasma monitored stability. Proteomic analysis identified upregulated proteins related to transport mechanisms epithelial/endothelial functions postprandially, indicating potential roles physiological responses nutritional intake. Enrichment analyses revealed vesicle-related pathways immune system processes. Flow cytometry showed increased expression CD324 on medium-sized CD9+CD63+CD81+ EVs suggesting an epithelial origin. These findings offer insights into lEV dynamics their significance, highlighting the need stringent guidelines EV studies account effects composition function.

Eosinophilic esophagitis (EoE) is a chronic inflammatory esophageal disorder. The lack of non-invasive biomarkers currently results in dependency on endoscopy with biopsies for its diagnosis and monitoring. We aimed to identify potential using microRNAs (miRNAs) plasma-derived extracellular vesicles (pEVs). This was prospective single-center observational study discovery cohort EoE patients (n = 26) active disease (EoE.Basal) after anti-inflammatory treatment (EoE.Post.tx) control subjects 16). Radiotherapy represents a major curative treatment for prostate cancer (PCa), but some patients will develop radioresistance (RR) and relapse. The underlying mechanisms remain poorly understood, miRNAs might be key players in the acquisition maintenance of RR. Through their encapsulation small extracellular vesicles (EVs), they can also relevant biomarkers radiation response. Using next-generation sequencing, we found that miR-200c-3p was downregulated PCa RR cells EVs due to gain methylation on its promoter during acquisition. We next showed exogenous overexpression restores radiosensitivity by delaying DNA repair through targeting HP1α. Interestingly, observed downregulation expression radiation-resistant lung breast cell lines. In summary, our study demonstrates could help distinguish radioresistant from sensitive tumor cells. This miRNA targets HP1α delay promote death.

Abstract Objective Patients with ovarian cancer (OC) may develop anti‐Yo‐associated paraneoplastic cerebellar degeneration (PCD)—a ataxia associated tumor‐induced autoimmunity against CDR2 and CDR2L proteins. Dysregulation of circulating exosomal microRNAs (miRNAs) occur in OC. Here, we investigated whether PCD is changes the miRNA profiles OC patients. Methods Serum exosomes were isolated from patients ( n = 15 ), 14 ) healthy controls (HC, ). Small RNA sequencing was used to identify differentially expressed miRNAs. Receiver operating characteristic curves evaluate biomarker sensitivity specificity, target prediction analysis employed elucidate gene targets. Results exhibited a distinct expression profile. We detected 103 miRNAs compared without 139 controls. Particularly miR‐486‐5p, miR‐4732‐5p, miR‐98‐5p miR‐21‐5p notable specificity for discriminating both showed that several targeted genes.
