Oxidative stress plays a critical role in various physiological and pathological processes, particularly during pregnancy, where it can significantly affect maternal fetal health. In the context of viral infections, such as those caused by Human Immunodeficiency Virus (HIV) severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), oxidative may exacerbate complications disrupting cellular function immune responses. Antiviral drugs, while essential managing these also contribute to stress, potentially impacting both mother developing fetus. Understanding mechanisms which antivirals examination pharmacokinetic changes pregnancy that influence drug metabolism is essential. Some research indicates antiretroviral drugs induce mitochondrial dysfunction other studies suggest their use generally safe. Therefore, concerns about long-term health effects persist. This review delves into complex interplay between antioxidant defenses, antiviral therapies, focusing on strategies mitigate potential damage. By addressing gaps our understanding, we highlight importance balancing efficacy with risks stress. Moreover, advocate for further develop safer, more effective therapeutic approaches pregnancy. dynamics optimizing outcomes fetus infections

The pursuit of a vaccine against the human cytomegalovirus (HCMV) has been ongoing for more than 50 years. HCMV is leading infectious cause birth defects, including damage to brain, and common complications in organ transplantation. complex biology made development difficult, but recent meeting sponsored by National Institute Allergy Infectious Diseases September 2023 brought together experts from academia, industry, federal agencies discuss progress field. reviewed status candidate vaccines under study challenges clinical trial design demonstrating efficacy congenital CMV infection or reduction disease following solid transplantation hematopoietic stem cell Discussion revealed that, with numerous that are study, it clear safe effective within reach. Cytomegalovirus infections and reactivations are more frequent in people living with HIV (PLWH) have been associated increased risk of progression immunosenescence. We explored the impact combination antiretroviral therapy (cART) on latent CMV infection 225 young adults parenterally infected during childhood. Anti-CMV IgG antibodies were present 93.7% participants, lower levels correlating longer cART exposure better immunologic parameters. Patients immunological treatment success (CD4 > 350 cells/mL) had significantly titers compared to those suboptimal immune response cART. In total, 78% tested patients robust CMV-specific T-cell responses, measured by an IFN-γ release assay. A good was cellular immunity: level positively correlated CD4 CD8-T cell counts. No differences observed between suppressed/non-suppressed viremia terms humoral response. DNA detected only 17% among cART-induced recovery. The successful subsequent reconstitution may lead restoration responses effective control infection, limiting episodes reactivation HIV-positive individuals.

Background/Objectives: Elevated cytokine levels, including IL-6 and IL-1β, can contribute to persistent brain inflammation in children with autism cytomegalovirus (CMV) infection, exacerbating autism-related behaviours symptoms. This study evaluates the impact of CMV-induced increases on eating sensory profiles autism. Methods: A cross-sectional design was employed, involving aged two five years (CMV-reactive IgG), ASD (n= 98) TD (n = 96). Serological tests using ELISA were conducted measure IgG CMV, IL-6, IL-1β biomarkers. Eating evaluated BAMBI (Brief Autism Mealtime Behaviour Inventory), assessed SSP (Short Sensory Profile). Statistical analyses performed Spearman’s rank chi-square tests. Results: The results show that significantly affects children’s (p < 0.001), notable differences found between groups. Correlation analysis revealed a significant association CMV 0.026) 0.014) group. Background: Cytomegalovirus (CMV) infection and age impact immune responses to vaccines. The effect of sex remains controversial. We investigated the relationship between cytomegalovirus-seropositivity, age, immunogenicity recombinant (RZV) live (ZVL) zoster vaccines in adults ≥50 years age. Methods: Varicella virus (VZV) glycoprotein E (gE)-specific antibody, antibody avidity, cell-mediated immunity (CMI) were measured pre-vaccination at regular intervals over 5 post-vaccination 80 RZV 79 ZVL recipients, including 91 cytomegalovirus-seropositive 90 female participants. Results: Differences associated with CMV-seropositivity: lower VZV-gE-CMI recipients after first dose vaccine, but no differences 2nd dose; VZV-gE-specific avidity recipients; more abundant Th1 senescent T cells (Tsen) less regulatory (Treg) tissue-resident memory (Trm). older age: cells. sex: none for either vaccine. cell subset abundance: higher Tsens Tregs or Trms post-dose 1 CMI Th1s concentrations. Conclusions: correlation CMV- age-associated subsets ZVLs RZVs suggests that imprinting contributes CMV on vaccine responses.

CMV is a ubiquitous DNA virus that establishes infection and results in 40–100% seropositivity. Viral replication occurs following an acquired primary (or reinfection) or by the reactivation of life-long latency. In immunocompetent patients, mostly asymptomatic mild self-limited. However, extensive review literature published up to April 2024 reveals despite immunocompetence, can cause very large variety clinical syndromes any part gastrointestinal tract (the most common pattern), central peripheral nervous system, eyes, as well hematological, pulmonary, cardiac, cutaneous disease. Not uncommonly, more than one system involved, though disease often self-limited, treatment with intravenous ganciclovir oral valganciclovir may be required, isolated cases, fatalities occur. Thus, potential should considered differential myriad non-immunocompromised patients. Associated systemic symptoms (fever, sweats, weight loss), lymphocytosis, hepatitis are not uncommon useful clue. Some populations, such critically ill patients intensive care, pregnant women, elderly those inflammatory bowel disease, susceptible. Moreover, past, latent (i.e., seropositivity) associated significant cardiovascular morbidity all-cause mortality years later intriguing requires further study. All these data indicate outstanding importance developing vaccine against CMV, which hopefully will become available foreseeable future. Meanwhile, solid diagnosis active quickly established ruled out) widely serology tests PCR amplification, clinicians all disciplines need aware diverse guises remember consider it host, including one.
