Alteration in gut microbiota has been associated with COVID-19. However, the underlying mechanisms remain poorly understood. Here, we outlined three potential interconnected mechanistic pathways leading to dysbiosis as an adverse outcome following SARS-CoV-2 presence gastrointestinal tract. Evidence from literature and current uncertainties are reported for each step of different pathways. One pathway investigates evidence that intestinal infection by inducing inflammation alters microbiota. Another links binding viral S protein angiotensin-converting enzyme 2 (ACE2) dysregulation this receptor, essential homeostasis—notably amino acid metabolism—leading dysbiosis. Additionally, could induce infecting bacteria. Assessing within Adverse Outcome Pathway framework justifies confidence proposed support disease management permits identification inconsistencies knowledge gaps orient further research.

The adverse outcome pathways (AOPs) were developed to accelerate evidence-based chemical risk assessment by leveraging data from new approach methodologies. Thanks their stressor-agnostic approach, AOPs seen as instrumental in other fields. Here, we present that report non-chemical stressors along with the challenges encountered for development. Challenges regarding linked nanomaterials include non-specific molecular initiating events, limited understanding of nanomaterial biodistribution, and needs adaptations silico modeling testing systems. It is known that the virus SARS-CoV-2 can attack gastrointestinal (GI) tract and induce gastroenteritis. This trigger a wide variety of disorders gut-brain interaction (DGBIs) or functional (FGIDs), including post-infectious dyspepsia, which remains underestimated. To estimate prevalence dyspeptic symptoms following COVID-19, immediately after discharge 3, 6, 9 months hospitalization.A prospective, single-centre evaluation questions regarding dyspepsia (FD) as assessed by Gastroduodenal Module ROME IV Diagnostic Questionnaire for Adult FGIDs among 320 patients who had COVID-19.The FD criteria were met at respective time-points 0.0% (0), 4.8% (12), 3.2% (8), (8) cases. However, presence GI suggested but did not meet timeframe found in 9.6% (24), 23.5% (59), 20.7% (52), (52) cases, respectively. The persistence COVID-19 significant problem. The Rome may underestimate number with persistent disease.

The current understanding of long COVID (LC) is still limited. This review highlights key findings regarding the role gut microbiota, mitochondria, and main pathophysiological aspects LC revealed by clinical studies, related to complex interplay between infection, intestinal dysbiosis, dysfunctional systemic inflammation generated in a vicious circle, reflecting molecular cellular processes from "leaky gut" electron transport chain (ETC)" into quantum leap. heterogeneity has hindered progress deciphering all mechanisms, therefore, approach must be multidisciplinary, with special focus not only on symptomatic management but also addressing underlying health problems patients. It imperative further assess validate effects COVID-19 microbiome their relationship infections other viral agents or pathogens. Further studies are needed better understand expand interdisciplinary points view that required accurately diagnose effectively treat this heterogeneous condition. Given ability SARS-CoV-2 induce autoimmunity susceptible patients, they should monitored for symptoms autoimmune disease after contracting infection. One question remains open, namely, whether various vaccines developed end pandemic will autoimmunity. Introduction The CIAO project was launched in Spring 2020 to address the need make sense of numerous and disparate data available on COVID-19 pathogenesis. Based a crowdsourcing model large-scale collaboration, has exploited Adverse Outcome Pathway (AOP) knowledge management framework built support chemical risk assessment driven by mechanistic understanding biological perturbations at different organizational levels. Hence AOPs might have real potential integrate produced through approaches from disciplines as experienced context COVID-19. In this study, we aim effectiveness AOP (i) supporting an interdisciplinary collaboration for viral disease (ii) working conceptual mediator collaboration. Methods We used survey disseminated among participants, workshop open all interested contributors, series interviews with some participants self-reflection processes. Results supported genuine interdisciplinarity exchange knowledge. provided common reference point discussion diagram assisted making explicit what are perspectives brought about pathways. AOP-Wiki showed up many aspects its usability those not already world AOPs. Meanwhile their use highlighted needed adaptations. new Wiki elements modulating factors potentially most disruptive one. ABSTRACT The aim of this study was to evaluate whether community-level monitoring respiratory and enteric viruses in wastewater can provide a comprehensive picture local virus circulation. Wastewater samples were collected weekly at the treatment plant (WWTP) inlet outlet nearby nursing home (NH) Burgundy, France, during winter period 2022/2023. We searched for pepper mild mottle as an indicator fecal content well main [severe acute syndrome coronavirus 2 (SARS-CoV-2), influenza, syncytial virus] (rotavirus, sapovirus, norovirus, astrovirus, adenovirus). Samples analyzed using real-time reverse transcription PCR-based methods. SARS-CoV-2 most frequently detected virus, with 66.7% positive from WWTP 28.6% NH. Peaks consistent chronological incidence infections recorded sentinel surveillance hospital databases. number lower NH than three viruses. Enteric detected, often sapovirus norovirus genogroup II, accounting both 77.8% 57.1% 37%, respectively, large circulation unexpected particular Combined simple optimized methods be valuable tool viral may serve suitable early warning system identifying outbreaks onset epidemics. These results encourage application wastewater-based (WBS) SARS-CoV2, sapovirus. IMPORTANCE WBS provides information on spread epidemic environment appropriate sensitive detection By PCR retirement (connected same collective sewer network), we aimed demonstrate that implementing combined key community sites allows effective occurrence (influenza, SARS-CoV-2) (norovirus, rotavirus, sapovirus) within given population. This analysis localized scale provided new two different sites. Implementing monitor or emergence infectious diseases is important means alerting authorities improving public health management. could participate actively humans, animals, environment.

ABSTRACT Many COVID-19 patients suffer from gastrointestinal symptoms and impaired intestinal barrier function may play a key role in Long COVID. Despite its importance, the impact of SARS-CoV-2 on epithelia is poorly understood. To address this, we established an model integrating epithelial Caco-2 cells, mucus-secreting HT29 cells human Raji cells. This gut allows efficient differentiation into microfold-like faithfully mimics function, highly permissive to infection. Early strains Delta variant replicated with high efficiency, severely disrupted depleted tight junction proteins, such as claudin-1, occludin ZO-1. In comparison, Omicron subvariants also ZO-1 junctions but had fewer damaging effects mucosal integrity function. Remdesivir TMPRSS2 inhibitor Camostat prevented replication thus damage, while Cathepsin E64d was ineffective. Our results support that disrupts further suggest circulating variants are less than earlier viral strains.
