Sodium-glucose cotransporter-2 (SGLT-2) inhibitors represent a cutting-edge class of oral antidiabetic therapeutics that operate through selective inhibition glucose reabsorption in proximal renal tubules, consequently augmenting urinary excretion and attenuating blood levels. Extensive clinical investigations have demonstrated their profound cardiovascular efficacy. Parallel basic science research has elucidated the mechanistic pathways which diverse SGLT-2 beneficially modulate pulmonary vascular cells arterial remodeling. Specifically, these exhibit promising potential enhancing endothelial cell function, suppressing smooth muscle proliferation migration, reversing remodeling, maintaining hemodynamic equilibrium. This comprehensive review synthesizes current literature to delineate mechanisms by enhance function reverse thereby offering novel therapeutic perspectives for diseases.

Der früher verwendete Begriff der kardiorenalen Krankheit ("cardiorenal disease", CRD) trifft die Komplexität zahlreichen Interaktionen zwischen Herz- und Niereninsuffizienz sehr viel besser als später Begrifflichkeit des oder kardiorenal-metabolischen Syndroms, wenngleich er CRD auch nur unzureichend gerecht wird. hiermit assoziierten Komorbiditäten sind pathophysiologisch, prognostisch nun therapeutisch eng miteinander verzahnt. Zunehmend rückt Stellenwert HFpEF ("heart failure with preserved ejection fraction") nicht sondern in den Vordergrund. Bisherige Konzepte wie sequenzielle Nephronblockade werden durch neuere, ineinandergreifende prognoseverbessernde z. B. dem panrenalen Diuretikatherapie abgelöst. Zudem führen neue systemübergreifende -modulierende innovative Therapieoptionen zu einem Paradigmenwechsel von organzentrierter Therapie, Symptomkontrolle -linderung hin zur tatsächlichen Prognoseverbesserung. Mit diesen neuen CAREPID prognosis improving drugs") stehen organübergreifende bei Verfügung.

Background:Sweet Tea (Lithocarpus polystachyus Rehd.), a traditional ethnobotanical medicine, contains phlorizin, dihydrochalcone compound with antioxidative and anti-inflammatory properties. Given the critical role of oxidative stress inflammation in age-related macular degeneration (AMD), this study tested hypothesis that phlorizin mitigates damage AMD models, thereby offering therapeutic potential. Materials Methods: Adult retinal pigmented epithelial cells (ARPE-19) were pre-treated (0.01–0.1 μM) subjected to induced by ultraviolet A (UVA) radiation or sodium iodate (NaIO3). Cell viability, reactive oxygen species (ROS) production, MAPK/NF-κB signaling, level pro-inflammatory cytokines (IL-1β, IL-6, TNF-α) pro-angiogenic factors (VEGF, MMP2, MMP9) expression assessed using MTT assays, fluorescence imaging, Western blotting, RT-qPCR. In vivo, laser-induced choroidal neovascularization (CNV) mouse model was used evaluate phlorizin’s effects on CNV formation vascular leakage via fundus photography angiography. Result: Phlorizin significantly enhanced cell reduced ROS inhibited activation, downregulated inflammatory angiogenic mediators. vivo studies confirmed following treatment. Conclusions: demonstrated significant protective against inflammation, highlighting its potential for treating AMD.

Among fruits, the apple is unique for producing large amounts of dihydrochalcone phloridzin, which, together with phloretin, its aglycone, valuable to pharmaceutical and food industries antidiabetic, antioxidant, anticarcinogenic properties, as well use a sweetener. We analysed phloridzin concentration, total phenolic content, antioxidant activity in peel, flesh, seeds, juice, pomace 13 international local varieties. In unprocessed fruit, seeds had highest while contents were mostly found peel. processed samples, compounds especially higher juice than pomace. Moreover, content was much content. Juice showed activity, followed by peel flesh. Across all did not directly correlate concentrations, suggesting that ascribed may need re-evaluation. Ferric Reducing Antioxidant Power (FRAP) assay, only at high concentrations when compared phloretin. Considering produced industry, residual promising source phloridzin. For technical use, processing this phloretin would be advantageous.
