Abstract Ageing as a natural irreversible process inherently results in the functional deterioration of numerous organ systems and tissues, including skeletal immune systems. Recent studies have elucidated intricate bidirectional interactions between these two In this review, we provide comprehensive synthesis molecular mechanisms cell ageing. We further discuss how age-related changes influence system consequent impact alterations on system. Finally, highlight clinical implications findings propose potential strategies to promote healthy ageing reduce pathologic both

Biotin (vitamin B7, or vitamin H) is a water-soluble B-vitamin that functions as cofactor for carboxylases, i.e., enzymes involved in the cellular metabolism of fatty acids and amino gluconeogenesis; moreover, reported, biotin may be gene regulation. not synthesized by human cells, but it found food also produced intestinal bacteria. status/homeostasis individuals depends on several factors, including efficiency/deficiency recycling within organism (biotinidase, holocarboxylase synthetase), and/or effectiveness uptake, which mainly accomplished through sodium-dependent multivitamin transporter. In last years, administration at high/"pharmacological" doses has been proposed to treat specific defects/deficiencies disorders, exhibiting neurological dermatological symptoms biotinidase deficiency, synthetase biotin-thiamine-responsive basal ganglia disease. On other hand, according warnings Food Drug Administration, USA, high levels can affect clinical biotin-(strept)avidin assays thus lead false results during quantification critical biomarkers. this review article, recent findings/advancements offer new insight abovementioned research fields concerning will presented briefly discussed.

High-grade serous ovarian cancer (HGSOC), the predominant subtype of epithelial cancer, is frequently diagnosed at an advanced stage due to its nonspecific early symptoms. Despite standard treatments, including cytoreductive surgery and platinum-based chemotherapy, significant improvements in survival have been limited. Understanding molecular mechanisms, immune landscape, drug sensitivity HGSOC crucial for developing more effective personalized therapies. This study integrates insights from immunology, profiling, analysis identify novel therapeutic targets improve treatment outcomes. Utilizing single-cell RNA sequencing (scRNA-seq), systematically examines tumor heterogeneity microenvironment, focusing on biomarkers influencing response activity, aiming enhance patient outcomes quality life. scRNA-seq data was obtained GEO database this study. Differential gene expression analyzed using ontology set enrichment methods. InferCNV identified malignant cells, while Monocle, Cytotrace, Slingshot software inferred differentiation trajectories. The CellChat package predicted cellular communication between cell subtypes other pySCENIC utilized transcription factor regulatory networks within subtypes. Finally, results were validated through functional experiments, a prognostic model developed assess prognosis, infiltration, across various risk groups. investigated scRNA-seq, their interactions microenvironment. We confirmed key role C2 IGF2+ HGSOC, which significantly associated with poor prognosis high levels chromosomal copy number variations. located terminal tumor, displaying higher degree malignancy close association IIIC tissue types. also metabolic pathways, such as glycolysis riboflavin metabolism, well programmed death processes. highlighted complex fibroblasts MK signaling pathway, may be closely related tumor-associated progression. Elevated PRRX1 connected impact disease progression by modulating transcription. A based demonstrated adverse outcomes, emphasizing importance infiltration clinical intervention strategies. oncology, immunotherapy, reveal mechanisms driving resistance. subtype, linked offers promising target future Emphasizing sensitivity, research highlights strategies life patients.

Parkinson's disease is recognized as the second most prevalent neurodegenerative disorder globally, with its incidence rate projected to increase alongside ongoing population growth. However, precise etiology of remains elusive. This article explores inflammatory mechanisms linking gut microbiota disease, emphasizing alterations in and their metabolites that influence disease's progression through bidirectional transmission signals along gut-brain axis. Building on this mechanistic framework, further discusses research methodologies treatment strategies focused metabolites, including metabolomics detection techniques, animal model investigations, therapeutic approaches such dietary interventions, probiotic treatments, fecal transplantation. Ultimately, aims elucidate relationship between underlying thereby paving way for novel avenues condition.
