Background/Objectives: Triple-negative breast cancer (TNBC) is the most challenging molecular subtype of (BC) in clinical practice, associated with a worse prognosis due to limited treatment strategies and its insensitivity conventional drugs. Zinc an important trace element for homeostasis, Schiff base metal complexes have shown promise treating advanced tumors. In this study, four new heteroleptic Zn(II) (1–4) bases were synthesized, characterized, evaluated their activity BC cells. Methods: Compounds crystal structures determined. Biological was assessed using MTT, clonogenic, scratch wound healing, caspase 3 8 activity, qPCR, chemosensitization assays. Results: The exhibited cytotoxicity against MCF-7 (luminal BC), MDA-MB-453 (HER2-positive MDA-MB-231 cell lines, IC50 values ranging from 0.01 20 µM. Complex 4 showed reduced toward non-tumor lines. This, complexation increased ligands, trend not observed 1–3. Due favorable profile, complex selected further assays, which it inhibited colony formation migration TNBC cells dose-dependent manner. Furthermore, compound induced death independently caspases, decreasing 8. Interestingly, sensitized TBNC doxorubicin paclitaxel, possibly modulating epithelial–mesenchymal transition mechanism, as evidenced by CDH1 expression. Conclusions: Results suggest potential sensitizing aggressive chemotherapy, proving be promising alternative cases therapeutic failure.

Breast cancer is one of the most common cancers worldwide, at same time being prevalent causes women’s death. Many factors such as alcohol, weight fluctuations, or hormonal replacement therapy can potentially contribute to breast development and progression. Another important factor in onset includes micronutrient status. In this narrative review, we analyzed 23 micronutrients their possible influence on Further, aim study was investigate impact status prevention its various therapeutic pathways. We researched meta-analyses, systemic reviews, retrospective studies, well original studies human animal models. The results these indicate a correlation between different levels decreased risk better survival rate. However, further are necessary establish adequate doses supplementation chosen exact mechanisms therapy.

An optimal amount of labile zinc (Zn 2+ ) is essential for proliferation human cells, where Zn levels that are too high or low cause cell cycle exit. Tumors the breast have been characterized by total . Given role in cells and elevation cancer tumors, we examined concentration across a panel 5 lines, compared to normal MCF10A line. We found three lines (MDA-MB-231, MDA-MB-157, SK-Br-3) showed elevated cytosol, while T-47D significantly lower , MCF7 no change cells. There was as measured ICP-MS, but did observe difference ability accumulate when media elevated. Therefore, how each line affected increases decreases media. striking differences, MCF7) robust at concentrations killed MCF10A, T-47D, SK-Br-3 also discovered 4 demonstrate compromised increased death suggesting these may be addicted Overall, our study suggests significant differences homeostasis regulation different types with consequences both viability.

Triple-negative breast cancer (TNBC) is usually the most malignant and aggressive mammary epithelial tumor characterized by lack of expression for estrogen receptors progesterone receptors, absence epidermal growth factor receptor (HER)2 amplification. Corresponding to 15–20% all cancers well-known its poor clinical outcome, this negative deprives TNBC from targeted therapy makes management therapeutically challenging. Type 2 diabetes mellitus (T2DM) common ageing metabolic disorder due insulin deficiency or resistance resulting in hyperglycemia, hyperinsulinemia, hyperlipidemia. Due hormonal imbalances, there are many interplays between both chronic disorders leading increased risk cancer, especially TNBC, diagnosed T2DM patients. The purpose review provide up-to-date information related epidemiology clinicopathological features, factors, diagnosis, biomarkers, current therapy/clinical trials patients with compared non-diabetic counterparts. Thus, in-depth investigation diabetic complications on onset, development, progression discovery biomarkers would improve through early tailoring a better outcome TNBC.

Abstract Background Dysregulation of iron metabolism has been shown to have significant implications for cancer development. We aimed investigate the prognostic and immunological significance metabolism-related genes (IMRGs) in nasopharyngeal carcinoma (NPC). Methods Multiple Gene Expression Omnibus (GEO) The Cancer Genome Atlas (TCGA) datasets were analyzed identify key IMRGs associated with prognosis. Additionally, was explored. Results A novel risk model established using LASSO regression algorithm, incorporating three (TFRC, SLC39A14, ATP6V0D1).This categorized patients into low high-risk groups, Kaplan–Meier analysis revealed significantly shorter progression-free survival group ( P < 0.0001). model’s accuracy additionally confirmed by employing time-dependent Receiver Operating Characteristic (ROC) curves conducting Decision Curve Analysis (DCA). High-risk found correlate advanced clinical stages, specific tumor microenvironment subtypes, distinct morphologies. ESTIMATE demonstrated a inverse relationship between increased immune, stromal, scores lowered score. Immune indicated negative correlation score abundance most tumor-infiltrating immune cells, including dendritic CD8 + T CD4 B cells. This extended checkpoint such as PDCD1, CTLA4, TIGIT, LAG3, BTLA. protein expression patterns selected NPC samples validated through immunohistochemistry. Conclusion study presents utilizing NPC, which could assist assessing patient prognosis provide insights new therapeutic targets NPC.

Background Breast cancer is one of the most serious and prevalent malignancies. Zinc commonly known to play a crucial role in development progression breast cancer; however, detailed mechanisms underlying this are not well understood. This study aimed develop zinc metabolism-related gene (ZMRG) signature based on multi-database predict patient prognosis investigate relationship between drug therapy response immune enrichment. Methods Data for samples from The Cancer Genome Atlas Gene Expression Omnibus databases were screened genes using Molecular Signature Database. Cox Least Absolute Shrinkage Selection Operator regressions performed construct ZMRG signature. To assess predictive performance signature, Kaplan–Meier analysis receiver operating characteristic curves used. Additionally, we utilised single-sample set enrichment analysis, Tumour Immune Estimation Resource, Genomics Drug Sensitivity database, Therapeutics Response Portal association tumour microenvironment sensitivity. Quantitative PCR was used expression each cell lines samples. Results Five ZMRGs identified (ATP7B, BGLAP, P2RX4, SLC39A11, TH) risk profile constructed each. Two groups, high- low-risk, way, high-risk score subgroups found have worse prognosis. validated GSE42568 dataset. sensitivity analyses showed that these five significantly associated with response. group substantial infiltration pathways, patients more sensitive drugs cancer.
