BACKGROUND: Flavonoids, a class of plant polyphenols, exhibit wide range biological (neuro- and immunotropic, antioxidant, anti-inflammatory, epigenome-modulating) properties involved in the mechanisms management various pathological processes, including nervous system diseases. Alcoholism is pervasive social, medical, economic issue modern society. Prolonged exposure to ethanol has direct mediated toxic effect on human body through its metabolites negatively affecting immune systems that play major role adaptation. The ability bioflavonoids manage disorders chronic diseases with neuroimmune pathogenesis by interacting specific cell surface receptors can provide therapeutic benefits alcoholism. AIM: To assess neurotropic immunomodulatory novel curcumin-based bioflavonoid composition during prolonged consumption. MATERIALS AND METHODS: content was measured aqueous-organic extracts using high-performance liquid chromatography (HPLC). Chronically alcoholized male (CBA×C57Bl/6)F1 mice who received 10% solution as sole source fluid six months were administered 30 days. Subsequent studies assessed alcohol motivation consumption free choice water, well behavioral parameters open field test, cytokine brain structures (prefrontal cortex, hypothalamus, hippocampus, striatum) enzyme immunoassay. intensity cellular humoral response determined severity delayed-type hypersensitivity relative number splenic antibody-forming cells, respectively. RESULTS: quantitative consisting curcumin, piperine, soybean isoflavonoids, epigallocatechin-3-gallate, triterpene saponins, β-carotene. Taking this context shown have positive expressed correcting alcoholism-related phenotype (reduced motivation, stimulation locomotor exploratory activity), accompanied decreased levels certain proinflammatory cytokines (most pronounced hippocampus). Stimulation also demonstrated after course treatment described composition. CONCLUSIONS: data support use an additional agent

Hericium erinaceus, commonly known as lion’s mane mushroom, has gained increasing scientific interest due to its rich composition of bioactive compounds and diverse health-promoting properties. This narrative review provides a comprehensive overview the nutritional therapeutic potential H. with particular focus on anti-inflammatory, antioxidant, antimicrobial activities. A structured literature search was performed using databases such PubMed, Scopus, Science Direct, Web Science, Google Scholar. Studies published in last two decades focusing erinaceus’ were included. The chemical erinaceus includes polysaccharides, terpenoids (hericenones erinacines), phenolic compounds, which exhibit potent antioxidant effects by scavenging reactive oxygen species (ROS) inducing endogenous enzymes. Additionally, shows promising activity against bacterial fungal pathogens, applications combating antibiotic-resistant infections. mushroom’s capacity stimulate nerve growth factor (NGF) synthesis highlighted preventing managing neurodegenerative diseases, Alzheimer’s Parkinson’s. Advances biotechnological methods, including optimized cultivation techniques novel extraction may further enhance bioavailability pharmacological erinaceus. Despite findings, clinical validation remains limited. Future research should prioritize large-scale trials, standardization elucidation pharmacokinetics facilitate integration into evidence-based medicine. functional food, nutraceutical, adjunct agent highlights need for interdisciplinary collaboration between researchers, clinicians, regulatory bodies.

Neurodegenerative diseases are among the major challenges in modern medicine, due to progressive aging of world population. Among these, Parkinson's disease (PD) affects 10 million people worldwide and is associated with aggregation presynaptic protein α-synuclein (α-syn). Here we use two different PD models, yeast cells neuroblastoma overexpressing α-syn, investigate protective effect an extract from cocoa shell, which a by-product roasting process beans. The LC-ESI-qTOF-MS NMR analyses allow identification amino acids (including essential ones), organic acids, lactate glycerol, confirming also presence methylxanthines, namely caffeine theobromine. present study demonstrates that supplementation bean shell (CBSE) strongly improves longevity expressing reducing level reactive oxygen species, activating autophagy intracellular aggresomes. These anti-aggregation properties confirmed cells, where CBSE treatment leads activation AMPK kinase significant reduction toxic α-syn oligomers. Results obtained by surface plasmon resonance (SPR) assay highlights binds concentration-dependent manner, supporting its inhibitory role on amyloid α-syn. findings suggest form nutraceuticals may represent promising way prevent neurodegenerative aggregation.
