Abstract Skin aging is influenced by structural alterations, oxidative stress, inflammation, and microbiome changes, a comprehensive approach to addressing these factors may be effective for mitigating skin aging. This study evaluates the multifaceted anti-aging effects of heat-killed (HK-HN910) lysed (LS-HN910) forms Lactobacillus paragasseri HN910. Protective on cell viability, permeability, nitric oxide (NO) production, gene expression both HK-HN910 LS-HN910 were observed. Both significantly enhanced tight junction (TJ) protein zonula occludens- 1 ( ZO- ) antioxidant enzyme glutathione peroxidase GPx) expression, while downregulating that senescence-associated secretory phenotype pro-inflammatory cytokines interleukin IL - 1α , IL- 1β 6 8 tumor necrosis factor-alpha TNFα ). showed greater upregulation GPx downregulation compared HK-HN910. Cell wall component D-alanine (D-Ala) was released in higher amounts than demonstrated effects. D-Ala upregulated barrier claudin- Cla- ), occludin OCC filaggrin FLG sphingomyelin phosphodiesterase 2 SMPD2 enzymes superoxide dismutase SOD catalase CAT . treatment clinically revealed improvements parameters, including transepidermal water loss, contents, sebum levels, dermal density, eye wrinkle index, pH, brightness, microbiota composition, with significant increase Rhodococcus abundance. These findings indicate LS-HN910, containing D-Ala, promising cosmeceutical preventing enhancing barrier, promoting defense, modulating inflammatory responses, influencing microbiota.
