Abstract Chronic migraine is one of the most common pain disorders, characterized by significant disability and a lack safe, long‐term, effective treatment options. Recent studies highlight interaction between lung microbiota central nervous system. In this study, nitroglycerin (NTG)‐induced chronic model constructed in male C57BL/6 mice to explore these interactions. Notable alterations are observed migraine‐afflicted mice. Notably, there marked decrease Proteobacteria group, associated with short‐chain fatty acids 5‐hydroxytryptamine (5‐HT). After intratracheal injection neomycin, diversity altered, resulting relief migraines.
