Abstract Background Cervical screening rates have fallen in recent years the UK, representing a health inequity for some under-served groups. Self-sampling alternatives to cervical may be useful where certain barriers prohibit access routine screening. However, there is limited evidence on whether self-sampling methods address known and subsequently increase uptake amongst under-screened Addressing this research gap, study aims understand experiences during attending groups and; explore views of individuals eligible towards (vaginal swabbing urine sampling) as alternative how existing Methods We draw three integrated theoretical frameworks (access primary care services, intersectional feminist perspectives) examine participants’ toward methods. undertook qualitative data collection (interviews focus groups) with 46 participants, facilitated by collaborations VCSE sector which successfully enhanced reach communities. Inflammation plays a crucial role in wound healing and the host immune response following pathogenic invasion. However, unresolved chronic inflammation can result tissue fibrosis genetic alterations that contribute to pathogenesis of human diseases such as cancer. Recent scientific advancements exploring underlying mechanisms malignant cellular transformations cancer progression have exposed significant disparities between pediatric adult-onset cancers. For instance, cancers tend lower mutational burdens arise actively developing tissues, where cell-cycle dysregulation leads gene, chromosomal, fusion gene development not seen counterparts. As such, findings adult cannot be directly applied cancers, unique mutations inherent etiologies remain poorly understood. Here, we review processes chromosomal instability, tumor microenvironment, tumorigenesis transformation explore current therapeutic interventions maintain and/or restore inflammatory homeostasis.

This comprehensive review encompasses studies examining changes in the cervical and cervico-vaginal microbiota (CM CVM) relation to human papillomavirus (HPV) using next-generation sequencing (NGS) technology. HPV infection remains a prominent global health concern, with spectrum of manifestations, from benign lesions life-threatening cancers. The CM CVM, unique collection microorganisms inhabiting cervix/vagina, has emerged as critical player health. Recent research indicated that disruptions characterized by decrease Lactobacillus overgrowth other bacteria, might increase risk persistence progression abnormalities. alteration or CVM been linked higher likelihood dysplasia. Long noncoding RNAs (lncRNAs) are RNA transcripts longer than 200 nucleotides that do not code for proteins but have been linked to cancer development and metastasis. Metastasis-associated lung adenocarcinoma transcript 1 (MALAT-1) influences crucial hallmarks through intricate molecular mechanisms, including proliferation, invasion, angiogenesis, apoptosis, the epithelial-mesenchymal transition (EMT). The current article highlights involvement of MALAT-1 in drug resistance, making it a potential target overcome chemotherapy refractoriness. It discusses impact on immunomodulatory molecules, such as major histocompatibility complex (MHC) PD-L1, leading immune evasion hindering anti-tumor responses. also plays significant role immunology by regulating diverse cell populations. In summary, is versatile regulator, influencing tumorigenesis, chemoresistance, immunotherapy Understanding its precise mechanisms developing targeted therapies, therapeutic strategies targeting show promise improving treatment outcomes. However, further research needed fully uncover biology translate these findings into clinical applications.

Cervical cancer continues to pose a significant health challenge, especially in resource-limited settings, highlighting the need for development of novel therapeutic agents. This study investigates potential 2,4-diphenyl indenol [1,2-b] pyridinol derivatives as inhibitors targeting epidermal growth factor receptor (EGFR) through computational drug discovery methods. A genetic algorithm-multiple linear regression (GA-MLR) model was created, achieving strong predictive accuracy with R² = 0.9243, Q² 0.8957, CCC 0.9021, and MAE 0.034. Molecular docking studies indicated that ligand 57 displayed highest binding affinity -29.2313 kcal/mol, followed by ligands 111 (-29.1459 kcal/mol) 110 (-29.9082 kcal/mol), all which stabilize key EGFR residues. dynamics (MD) simulations confirmed stability 111, showing an improved free energy -18.2235 kcal/mol. Additionally, pharmacokinetic analysis further validated their favorable ADMET (Absorption, Distribution, Metabolism, Excretion, Toxicity) properties, supporting drug-like candidates. These findings establish foundation EGFR-targeted therapies cervical cancer.

Vaginal dysbiosis is characterized by a decrease in the relative abundance of Lactobacillus species favor other species. This condition facilitates infections sexually transmitted pathogens including high risk (HR)-human papilloma viruses (HPVs) involved development cervical cancer. Some vaginal bacteria contribute to neoplastic progression inducing chronic inflammation and directly activating molecular pathways carcinogenesis. In this study, SiHa cells, an HPV-16-transformed epithelial cell line, were exposed different representative microbial communities. The expression HPV oncogenes E6 E7 production oncoproteins was evaluated. results showed that crispatus gasseri modulated basal genes cells oncoproteins. had contrasting effects on E6/E7 gene protein production. increased strains Gardnerella vaginalis and, lesser extent, Megasphaera micronuciformis. contrast, Prevotella bivia decreased protein. A amount p53 pRb found cultures with M. micronuciformis, accordingly, same cultures, higher percentage progressed S-phase cycle compared untreated or Lactobacillus-stimulated cultures. These data confirm L. represents most protective component microbiota against HR-HPV infected while micronuciformis G. may interfere oncogenic process, maintaining viral
