Ergosterol plays a dual role in fungal pathogenesis and azole resistance, driving key advancements the understanding of its biosynthesis regulation. This review integrates latest research progress on regulation ergosterol drug resistance pathogenicity. We comprehensively discuss functions enzymes (such as Erg11p/Cyp51A, Erg6p, Erg3p, Erg25p) mevalonate, late, alternative pathways. Notably, we highlight complex cyp51A expression by factors such SrbA, AtrR, CBC, HapX, NCT Aspergillus fumigatus, elucidate distinctive roles Upc2, Adr1, Rpn4 Candida species. Importantly, summarize recent discoveries CprA-dependent Cyp51A/Erg11p heme-mediated stability control. Based these findings, propose innovative antifungal strategies, including dual-target inhibition multi-enzyme natural products, which provide novel insights potential directions for development next-generation therapies.
