The unique anatomical and physiological connections between the nasal cavity brain provide a pathway for bypassing blood–brain barrier to allow direct brain-targeted drug delivery through administration. There are several advantages of administration compared with other routes; example, first-pass effect that leads metabolism orally administered drugs can be bypassed, poor compliance associated injections minimized. Nasal also help maximize delivery, allowing high pharmacological activity at lower dosages, thereby minimizing likelihood adverse effects providing highly promising treatment central nervous system diseases. aim this review article was briefly describe structures brain, pathways which enter nose, factors affecting methods improve systems application related biomaterials, common experimental used in intranasal research, current limitations such approaches, solid foundation further in-depth research on (see Graphical Abstract ).

Alzheimer's disease, a progressive neurodegenerative condition, is characterized by gradual decline in cognitive functions. Current treatment approaches primarily involve the administration of medications through oral, parenteral, and transdermal routes, aiming to improve function alleviate symptoms. However, these treatments face limitations, such as low bioavailability inadequate permeation. Alternative invasive methods, while explored, often entail discomfort require specialized assistance. Therefore, development non-invasive efficient delivery system crucial. Intranasal has emerged potential solution, although it constrained unique conditions nasal cavity. An innovative approach involves use nano-carriers based on nanotechnology for intranasal delivery. This strategy overcome current limitations providing enhanced bioavailability, improved permeation, effective traversal blood-brain barrier, extended retention within body, precise targeting brain. The comprehensive review focuses advancements designing various types nano-carriers, including polymeric nanoparticles, metal lipid liposomes, nanoemulsions, Quantum dots, dendrimers. These are specifically tailored therapeutic agents aimed at combatting disease. In summary, utilization systems show significant surmounting constraints disease strategies. Nevertheless, essential acknowledge regulatory well toxicity concerns associated with this route; meticulous consideration required when engineering carrier. underscores revolutionize management highlights importance addressing considerations safe implementations. Embracing could lead substantial field treatment.

Topical and transdermal drug delivery are advantageous administration routes, especially when treating diseases conditions with a skin etiology. Nevertheless, conventional dosage forms often lead to low therapeutic efficacy, safety issues, patient noncompliance. To tackle these novel topical platforms involving nanotechnology have been developed. This review focuses on the latest advances regarding development of nanoemulgels for application, encapsulating wide variety molecules, including already marketed drugs (miconazole, ketoconazole, fusidic acid, imiquimod, meloxicam), repurposed (atorvastatin, omeprazole, leflunomide), natural-derived compounds (eucalyptol, naringenin, thymoquinone, curcumin, chrysin, brucine, capsaicin), other synthetic molecules (ebselen, tocotrienols, retinyl palmitate), wound healing, appendage infections, inflammatory diseases, cancer, neuropathy, or anti-aging purposes. Developed formulations revealed adequate droplet size, PDI, viscosity, spreadability, pH, stability, release, permeation and/or retention capacity, having more characteristics than current formulations. In vitro in vivo studies established efficacy developed formulations, confirming their potential, making them promising replacement therapies, as possible adjuvant treatments, which might someday effectively reach market help fight highly incident systemic conditions.

Neuropsychiatric disorders are multifaceted syndromes with confounding neurological explanations. It includes anxiety, depression, autism spectrum disorder, attention deficit hyperactivity schizophrenia, Tourette’s syndrome, delirium, dementia, vascular cognitive impairment, and apathy etc. Globally, these occupy 15% of all diseases. As per the WHO, India has one largest populations people mental illnesses worldwide. The blood-brain barrier (BBB) makes it extremely difficult to distribute medicine target cells in brain tissues. However, is possible through novel advancements nanotechnology, molecular biology, neurosciences. One such cutting-edge delivery method, nose-to-brain (N2B) drug using nanoformulation (NF), overcomes traditional formulation limitations. Later offers more controlled release, better bioavailability, improved patient acceptance, reduced biological interference, circumvention BBB. When medicines delivered via intranasal (IN) route, they enter nasal cavity go connections between olfactory trigeminal nerves mucosa N2B. Delivering phytochemical, bioactive synthetic NF being investigated N2B strategy. mucociliary clearance, enzyme degradation, translocations by efflux mechanisms significant issues associated delivery. This review article discusses types neuropsychiatric their treatment plant-derived as well drug-loaded NFs administered IN-delivery system. In conclusion, this provided a comprehensive critical overview IN applicability for psychiatric disorders.

Depression and anxiety are high incidence debilitating psychiatric disorders, usually treated by antidepressant or anxiolytic drug administration, respectively. Nevertheless, treatment is given through the oral route, but low permeability of blood–brain barrier reduces amount that will be able to reach it, thus consequently reducing therapeutic efficacy. Which why it imperative find new solutions make these treatments more effective, safer, faster. To overcome this obstacle, three main strategies have been used improve brain targeting: intranasal route which allows directly transported neuronal pathways, bypassing avoiding hepatic gastrointestinal metabolism; use nanosystems for encapsulation, including polymeric lipidic nanoparticles, nanometric emulsions, nanogels; molecule functionalization ligand attachment, such as peptides polymers. Pharmacokinetic pharmacodynamic in vivo studies’ results shown administration can efficient targeting than other routes, nanoformulations quite advantageous increasing brain–drug bioavailability. These could key future improved therapies depressive disorders.
