The fusion of penetrating peptides (PPs), e.g., cell penetration (CPPs) or antimicrobial (AMPs), together with agents is an expanding research field. Specific AMPs, such as lactoferricin B (LfcinB), have demonstrated strong antibacterial, antifungal, and antiparasitic activity, well valuable anticancer proving beneficial in the development conjugates. resulting conjugates offer potential dual functionality, acting both agent. This especially necessary cancer treatment, where microbial infections pose a critical risk. Leukemic cells frequently exhibit altered outer lipid membranes compared to healthy cells, making them more sensitive compounds that interfere their membrane. In this study, we revisited reanalyzed our earlier on LfcinB its Furthermore, carried out new experiments specific focus proliferation, changes membrane asymmetric phosphatidylserine location, intracellular reactive oxygen species (ROS) generation, mitochondrial functions, vitro bacterial topoisomerase inhibition.

Abstract The study was conducted to analyze the antimicrobial activity and sensitivity of bovine lactoferrin against foodborne pathogens evaluate pH range thermal stability lactoferrin. agar well diffusion test used check each pathogen. Bacterial samples were purified confirmed by biochemical tests. Three concentrations 0.5, 1, 10 mg/ml evaluated for antibacterial Escherichia coli, Salmonella enteritidis, Staphylococcus aureus. Our results indicated that all three pathogenic bacteria inhibited, even at 0.5 (p < .05). E. coli most sensitive, S. aureus least sensitive. Lactoferrin remains effective 6, 7, 8, 9 pH, but no inhibition showed 3, 4, 5 change a pasteurization temperature 60–75 °C .05) inactivated higher temperatures. can inhibit from growth remain unchanged ranges 6–9.

Tumor necrosis factor (TNF) receptor 1 (TNFR1) plays a pivotal role in mediating TNF induced downstream signaling and regulating inflammatory response. Recent studies have suggested that TNFR1 activation involves conformational rearrangements of preligand assembled dimers targeting dynamics is viable strategy to modulate signaling. Here, we used combination biophysical, biochemical, cellular assays, as well molecular simulation show an anti-inflammatory peptide (FKCRRWQWRMKK), which termed FKC, inhibits allosterically by altering the states dimer without blocking receptor–ligand interaction or disrupting dimerization. We also demonstrated efficacy FKC showing HEK293 cells attenuates inflammation mice with intraperitoneal injection. Mechanistically, found binds cysteine-rich domains (CRD2/3) perturbs required for activation. Importantly, increases frequency opening both CRD2/3 CRD4 dimer, induces cytosolic regions receptor. This results inhibitory state impedes recruitment molecules. Together, these data provide evidence on feasibility conformationally active region open new avenues receptor-specific inhibition

Emerging infectious diseases are caused by unpredictable viruses with the dangerous potential to trigger global pandemics. These typically initiate infection utilizing anionic structures of host cell surface receptors gain entry. Lactoferrin (Lf) is a multifunctional glycoprotein multiple properties such as antiviral, anti-inflammatory and antioxidant activities. Due its cationic structure, Lf naturally interacts certain receptors, heparan sulfate proteoglycans, well viral particles other that targeted viruses. Therefore, may interfere virus-host interactions acting receptor competitor for Herein we summarize studies in which this competition was investigated SARS-CoV-2, Zika, Dengue, Hepatitis Influenza vitro . have demonstrated not only Lf’s competitive properties, but also intracellular impact on cells, enhancing survival reducing efficiency inhibiting enzymes. In addition, immunomodulatory effect highlighted, it can influence activity specific immune cells regulate cytokine release, thereby host’s response infections. Collectively, these promote promising candidate research diseases.

Lactoferrin (LF), a multifunctional glycoprotein with high iron-binding affinity, plays critical role in modulating physiological processes through its ability to reversibly bind and release iron ions, existing two distinct states: iron-saturated (holo-LF, > 85% saturation) iron-deficient (apo-LF, < 5% saturation). However, the importance of saturation has been largely overlooked LF production research due lack standardized protocols. The level dictates functional specificity: apo-LF exhibits potent antimicrobial properties by chelating disrupting membrane integrity, while also significantly inhibiting oxidative stress, thereby alleviating neurological disorders immune responses. In contrast, holo-LF participates metabolism transport, influencing tumor cell proliferation systemic uptake. This review systematically evaluates interplay between levels LF's biological functions, emphasizing dual roles human homeostasis disease modulation. Future should prioritize elucidating mechanisms underlying saturation-dependent bioactivity metabolic differences, incorporating emerging technologies enhance stability refine measurement accuracy.
