The present study aims to investigate the potential of 3D printing technique design gastroretentive floating tablets (GFTs) for modifying drug release profile an immediate-release tablet. A 3D-printed shell enclosing a captopril tablet was designed having varying number drug-release windows. impact geometrical changes in delivery system and thermal cross-linking polymers were evaluated observe influence on ability release. Water uptake, water insolubilization, Differential Scanning Calorimetry (DSC), Attenuated Total Reflection-Fourier Transform Infrared Spectroscopy (ATR-FTIR) performed assess degree polyvinyl alcohol (PVA) filament. GFT9 considered optimized gastric that exhibited >12 hours total time with zero lag successfully accomplished modified-drug by exhibiting >80% 8 hours. zero-order model, r2 value 0.9923, best fitted kinetic data GFT9, which followed super case II transport mechanism n 0.95. device (GFT9) also highest MDT values (238.55), representing slow from due crosslinking presence single drug-releasing window device.

Gastric acid secretion is closely associated with the development and treatment of chronic gastritis, gastric ulcers, reflux esophagitis. However, affected by complex physiological pathological factors, real-time detection control are complicated expensive. A delivery system for antacids therapeutics in response to low pH stomach holds promise smart personalized diseases. In this study, pH-responsive modular units were used assemble various devices self-regulation drug stomach. The unit a release window 50 mm2 could respond self-regulate within 10 min, which related its downward floatation internal gas production. assembled stably float medium detach sequentially at specific times. loaded exhibited under conditions. addition, suppressors multi-pulse or prolong after rapid neutralization acid. Compared traditional coating technology, 3D printing can print shell layer layer, flexibly adjust external structure composition, it into multi-level system. coating, 3D-printed shells have advantage flexible adjustment easy This provides universal strategy diseases abnormal secretion, especially delivering acid-unstable drugs.

Currently, fused deposition modeling (FDM) is a 3D printing technology that has been most widely used to develop innovative drug delivery approaches for overcoming the limitations of oral administration. Propranolol short plasma half-life and well soluble in acidic environments. Thus, this study aimed gastro-floating printed device (GFD) sustain release propranolol stomach as gastro-retentive system. The polylactic acid (PLA) was selected fabricate GFD. An air chamber included interior construction GFD design buoyancy. number open channels on side wall modified regulate release. gel formulation composed mixture polyvinylpyrrolidone (PVP) at weight ratio 6:5 then loaded into GFDs using syringe. exhibited floating ability more than 24 h with low standard deviation (SD) values variation shape dimension. from shows sustained properties simulated gastric environment without lag time. 4 5 6 h.
