Abstract Cadmium (Cd) is a global pollutant, and its accumulation in the liver causes oxidative stress, inflammation, insulin resistance (IR), metabolic complications. This study investigated whether curcumin treatment could alleviate hepatic IR Wistar rats exposed to sub-chronic cadmium explored underlying molecular pathways. Male were divided into control group (standard normocaloric diet + cadmium-free water) drinking water with 32.5 ppm CdCl 2 ) for 30 days. Oral glucose tolerance, response, assessed using mathematical models. Liver tissue was analyzed markers of key regulatory pathways, including NF-κB, Nrf2, MAPKs (JNK p38), IRS1-Akt pathway. We established an effective dose 250 mg/kg 5 days orally. Results demonstrated that after exposure, accumulated liver, inducing inflammatory state. characterized by increased expression JNK, p38, along diminished Nrf2 expression, IR, hyperglycemia, hyperinsulinemia. Curcumin effectively alleviated these disorders restoring balance between NF-κB modulating MAPK pathway, and, consequently, improving balance. In conclusion, this suggests induces through imbalance signaling appears improve thereby ameliorating IR.
