Zinc finger proteins (ZFP) are metalloproteins whose zinc atom interacts with side chains of specific histidines and cysteines the motif, generating functional three dimensional structures. CCCH zinc finger proteins (ZFPs) function mainly as RNA-binding (RBPs) and play a central role in the mRNA metabolism. Over twenty seven CCCH-ZFPs are encoded genome of human malaria parasite Plasmodium falciparum, causative agent tropica. However, little is known about their functions. In this study, we characterize one member PfCCCH-ZFP named ZNF4. We show that ZNF4 highly expressed mature gametocytes, where it predominantly localizes to cytoplasm. Targeted gene disruption showed no significant effect asexual blood stage replication gametocyte development while male exflagellation was significantly impaired, leading reduced transmission mosquito. Comparative transcriptomics between wildtype (WT) ZNF4-deficient line (ZNF4-KO) demonstrated deregulation 473 genes (274 upregulated 199 downregulated) gametocytes. Most downregulated peak expression with enriched associated axonemal dynein complex formation, cell projection organization affected, pointing phenotype exflagellation. Upregulated ATP synthesis. Our combined data therefore indicate protein which plays an important through regulation gametocyte-enriched genes.
